Objective:We used illumina high-throughput sequencing to analyze the intestinal flora and reveal the process of establishment of intestinal flora in infants with different feeding mode,so as to provide theoretical support for the research and development of formula.Methods:Twelve normal babies born at the Jilin University First Hospital were admitted into the study.Divided into standard infant formula-fed group,standard infant formula-fed with L.paracasei ssp.paracasei strain F19 group.Stool samples were collected on day 14,2 months,4 months,6months,8months,10 months,12months after birth,and were allocated to groups A0 to A12,B0 to B12,and stored in a-80℃ refrigerator.A high-throughput sequencing platform was used to sequence the V3,V4 variable area.The diversity and abundance of intestinal flora were analyzed and identified in their respective phylum,class,order,family,genus.Results:(1)The stool samples from the formula-fed group were covered 100% of the microflora,namely: Veillonella.formula-fed group were found 645 kinds of bacteria.The main advantage Intestinal flora: At the phylum level,the main advantageous bacteria were Firmicutes,Proteobacteria.At the class level,the main advantageous bacteria were Gammaproteobacteria,Bacteroidia.At the order level,the main advantageous bacteria were Enterobacteriales,Bacteroidales.At the family level,the main advantageous bacteria were Enterobacteriaceae,Veillonellaceae.At the genus level,the main advantageous bacteria were Veillonella,Bacteroides.(2)The stool samples from the formula-fed with L.paracasei ssp.paracasei strain F19 group were covered 100% of the microflora,namely: Streptococcus,Veillonella,Escherichia/Shigella.formula-fed with L.paracasei ssp.paracasei strain F19 group were found 793 kinds of bacteria.The main advantage Intestinal flora: At the phylum level,the main advantageous bacteria were Firmicutes,Proteobacteria.At the class level,the main advantageous bacteria were Gammaproteobacteria,Clostridia.At the order level,the main advantageous bacteria were Enterobacteriales,Clostridiales.At the family level,the main advantageous bacteria were Enterobacteriaceae,Veillonellaceae.At the genus level,the main advantageous bacteria were Bifidobacterium,Veillonella.(3)The changes of intestinal flora with formula fed: Bifidobacterium gradualy increased from birth to eight months,12 months have a descending trend.Escherichia/Shigella,Clostridium increased from birth to fourth months,12 months to decline.(4)The changes of infant intestinal flora formula-fed with L.paracasei ssp.paracasei strain F19 group : Bifidobacterium gradualy increased from birth to twelve months.Escherichia/Shigella increased from birth to four months,6 months have a descending trend,12 months to rise again.Clostridium declined from birth to fourth months,8months to rise,12 months have a descending trend.Conclusion:(1)The diversity of infant intestinal flora with probiotic F19 formula fed was higher than that of infants with formula fed.The relative abundance of Bifidobacteria in infant intestinal flora with probiotic F19 formula fed was significantly higher than that of infants with formula fed.L.paracasei ssp.paracasei strain F19 may be promote the reproduction of Bifidobacteria.(2)The diversity of infant intestinal flora was high,but there was no significant difference in intestinal flora diversity between formula fed and formula with probiotics F19 fed(P<0.05).(3)The dominant flora of formula fed group and formula with probiotics F19 fed were similar. |
