The quinolones are synthetic antibacterial drugs developed rapidly in recent years.The mechanism of action is to inhibit the synthesis of bacterial DNA,which leads to the degradation of DNA and the death of bacteria.The appearance of quinolones created a new era of synthetic antibiotics.At first,the drugs are used in the treatment of urinary tract infections,and now it has been developed to treat the system diseases of infectious,and widely used in animal husbandry as the prevention and treatment drugs.In recent years,the residues of the drugs in animal tissues have been attracted widespread attention.This paper studied the separation effects of six quinolones on the twospecification C18 liquid chromatography columns of the Diamonsil C18(2)(250 4.6mm * 5 m)and ZORBAX Extend C18(250mm * 3.0mm,5 m);studied the effects of sensitivity of six quinolones on the two UV detection wavelength of 254 nm and 280nm;studied the effects of recoveries of six quinolones on eight sample pretreatment methods;studies the effects of recoveries of six quinolones on four kinds of mobile phases and the effects of separation of six quinolones on the mobile phase ratio.This paper established the determination multi-residue for the six quinolones of danofloxacin,ofloxacin,enrofloxacin,ciprofloxacin,norfloxacin and Sarafloxacin by high performance liquid chromatography with ultraviolet detector.Samples extracted by organic solvent acetonitrile,centrifugation,defatted by hexane,purified by PXA SPE column.Linear range of the method is 0.01 ~ 5.00mg/L.The detection limit of instrument:ofloxacin is 0.0032g/mL;norfloxacin is 0.0012g/mL;ciprofloxacin is0.0018g/mL;enrofloxacin is 0.0020g/mL;danofloxacin is 0.0015g/mL;Sarafloxacin is 0.0019g/mL.The detection limit of method: ofloxacin is0.0303g/g;norfloxacin is 0.0385g/g;ciprofloxacin is 0.0330g/g;enrofloxacin si 0.0345g/g;danofloxacin is 0.0304g/g;Sarafloxacin is 0.0334g/g.Precision:ofloxacin is 4.93%~9.59%;norfloxacin is 5.03%~12.82%;ciprofloxacin is3.37%~13.73%;enrofloxacin is 12.01%~14.72%;danofloxacin is10.62%~16.03%;Sarafloxacin is 12.01%~12.98%.The recovery rates:ofloxacin is 100.7%~112.9%;norfloxacin is 100.7%~105.5%;Ciprofloxacin is 98.6%~108.7%;enrofloxacin is 104.5%~107.8%;danofloxacin is82.2%~99.6%;Sarafloxacin is 95.1%~111.7%.Afer comparison with the quinolones residues of the State regulations,the determination of quinolones residues in the animal origin food by high performance liquid chromatography with ultravioletwave detector has a certain practical value,is suitable for basic level laboratories.
