After removal of the caries and diseased teeth,the alveolar ridge will undergo absorption and atrophy,resulting in the loss of important soft and hard tissues.Periodontal disease can also cause atrophy and defect of the alveolar ridge.Insufficient amount of alveolar bone can lead to subsequent failure to perform effective dental implant restoration.In order to control the alveolar ridge absorption,it is necessary to preserve the alveolar fossa or alveolar ridge to avoid insufficient alveolar ridges in the future for traditional restoration or implant restoration.A method of implanting a guided bone regeneration(GBR)membrane is currently more commonly used.However,obtaining GBR with sufficient mechanical strength,predictable degradation rate,mimicking a structure similar to that of natural extracellular matrix(ECM)remains problematic.Natural polymer membranes have excellent biological properties,such as cell adhesion and biodegradation,but generally lacking sufficient mechanical strength and degradation curves;synthetic polymer membranes generally have better mechanical strength and poor biological activity.Therefore,blending natural polymers with synthetic polymers can not only compensate for the defects of the materials,but also has both materials' properties.In this paper,silk fibroin(SF)and poly-L-lactide caprolactone(PLCL)were used to prepare a blended GBR membrane by electrospinning.Firstly,the SF/PLCL single-layered electrospun membranes were prepared and their structure and properties were studied.Based on the multi-functional comprehensive requirements for GBR membranes,this project also prepared a bi-layered membranes.The outer layer acts as a support which is fabricated by SF/PLCL electrospun membrane wrapped with casted silk fibroin membrane.The inner layer is an SF/PLCL electrospun membrane,and its porous structure is beneficial to cell adhesion and proliferation.Futher,the morphology,mechanical and surface properties of the bi-layered membrane were studied.In addition,the drug release behavior and cytocompatibility of the bi-layered membrane loaded with drugs of promoting bone regeneration were also studied.Results showed that the structure of the electrospun membrane is influenced by SF/PLCL ratio,the morphology and mechanical properties of the electrospun membrane can be controlled by the proportion of the components.In addition,the mechanical properties of SF/PLCL bi-layered membrane could meet clinical requirements.The surface of the sample was hydrophilic,and the drug-loaded bi-layered membrane has good cell compatibility and of potential for promoting bone regeneration.It indicates that the membrane prepared in this paper have potential application value in the field of stomatology.
