| Soybean-based infant formula powder has high nutritional value,but its raw material,soybean,is recognized as one of the eight major food allergens.The allergic reaction caused by soybean is harmful to human health.Enzymatic hydrolysis is considered to be an effective method in reducing food allergenicity,and soybean sprouts processed from soybean can significantly improve their nutritional value and utilization rate of nutrients.Therefore,the enzymatic hydrolysis of protein in soybean sprout can destroys the allergen epitope and reduces its allergenicity.Thus,hypoallergenic soy-based infant formula powder prepared from the hydrolysate of soybean sprout protein can enrich the variety of infant formula powder and solve the problems of infant’s allergy to milk protein and lactose intolerance.It is of great significance for the development of hypoallergenic formula food.In this paper,hypoallergenic soybean peptides were used as raw materials to prepare hypoallergenic soy-based infant formula powder according to wet mixing process.The sensory,physicochemical properties,nutritional value and functional characteristics of homemade soy-based infant formula powder were evaluated.The digestibility of soy-based infant formula powder and the immunoreactivity of digestion products were evaluated.The potential allergenicity of soy-based infant formula and its digestion products were evaluated by using KU812 cells as a model.And factory design was done for hypo-allergenic soy-based formula powder.The main methods,results and conclusions of the study are as follows.1.The soybean peptide,a soybean sprouts proteolytic product,was used to prepare hypoallergenic soy-based infant formula powder by adding other nutrients,mixing,homogenizing,sterilizing and spray drying.The sensory index,physicochemical properties,nutritional value and functional evaluation of the homemade soy-based infant formula powder were carried out,and the commercially available infant formula powder was used as a reference group.The results showed that the sensory index and some physicochemical properties of homemade soy-based infant formula meet the requirements of GB 10765-2010"Food Safety National Standard Infant Formula Food",in which the content of moisture,protein and amino acid were higher than those of the control group,and the crude fat content is lower that of the control group.In nutritional value evaluation,the chemical score,amino acid score,essential amino acid index,biological value and nutritional index of self-made hypoallergenic soy-based infant formula powder(enzymatic soybean sprout protein)were higher than those of the control group,and the soy-based infant formula powder made of soybean sprout protein had the highest nutritional value.In the functional evaluation,the self-made hypoallergenic soy-based infant formula powder has poor wettability and dispersibility compared with the commercially available soy-based infant formula powder,but the emulsifying property,emulsion stability,foaming property,foaming stability and solubility were higher than those of the control group.2.The soy-based infant formula was digested in vitro simulation of infantile gastrointestinal tract,and their digestibility was assessed by protein digestibility,SDS-PAGE and Tricine-SDS-PAGE.The indirect competitive ELISA was used to evaluate the immunoreactivity of the digestive products.The results showed that the highest digestibility of the enzymatically treated soy-based infant formula in stomach and intestine was 58.25%and 71.39%,respectively.The electrophoresis analysis suggested that the enzymatically treated soy-based infant formula had the highest degree of degradation.While the results of the immunoreactivity showed that the IgG and IgE binding ability of the digestive products was the lowest.3.To evaluate the potential allergenicity of soy-based infant formula powders,the bioactive mediators(histamine,β-HEX and TNF-α)and intracellular Ca2+released by basophil KU812 activation degranulation were detected.The results showed that the KU812 cells stimulated by the enzymatic hydrolysis of soy-based infant formula powder had the lowest degree of degranulation,and its potential allergenicity was the lowest.Compared with the commercially available soy-based infant formula,the intracellular Ca2+content decreased by 34.93%,the histamine content decreased by5.86%,theβ-HEX release rate decreased by 17.03%,and the TNF-αrelease decreased by 36.69%.4.The factory design of hypoallergenic soy-based infant formula powder with an annual output of 600 tons was carried out in this paper,including the general layout,production process and technical and economic analysis.The plant area plan,equipment flow chart and process flow chart were also drawn.The annual output of600 tons of hypoallergenic soy-based infant formula powder was calculated,with annual sales of 75 million yuan and annual net profit of 12.53 million yuan. |
