Peony seed oil is a kind of edible woody vegetable oil rich in unsaturated fatty acids.In 2011,the peony oil food certification through the national new resources,more and more people’s attention to the peony oil,but less research on oil by-product peony seed meal,peony seed meal contains abundant phospholipids,polysaccharide,protein,dietary fiber,etc.,reasonable use of these components is of great significance on improving comprehensive utilization of peony seed.This study,according to the peony seed research and using status in shandong heze peony seed meal after seed oil as raw material,the extraction and purification of peony phospholipids,phospholipids of amphiphilic molecule,dihydro quercetin has many biological activities,but its worse through biological membrane,biological utilization is low,the amphiphilic phospholipids and dihydro quercetin compound,can improve the membrane permeability of dihydro quercetin,control their release.The main research contents and conclusions are as follows:1.The contents of fat,water,ash,total dietary fiber,protein,crude polysaccharide and phosphorus were determined to be 6.6%,1.56%,4%,4.64%,35.97%,6.6%and 1.35%,respectively.In addition,trace elements and other chemical components in peony seed meal accounted for 6.76%.2.N-hexane after removing the peony seed meal residual oil at low temperature,using ultrasonic-microwave method to extract the peony seed meal of phospholipids,by single factor and response surface method to optimize the best extraction process of peony seed meal phosphatide is:90%ethanol concentration,microwave time,180 s,microwave power is 800 w,and solid-liquid ratio is 1:6,extract four times,ultrasonic power fixed 50 w.Under the conditions of this process,the extraction rate of phospholipid was 78.38%,and the concentrated phospholipid of peony was obtained by the concentration and drying of ethanol extract,among which the content of phospholipid was 84.02%.3.Peony phospholipid ultrasonic dispersion in acetone,peony phospholipids by acetone insolubles refined purification,purification conditions as follows:the peony phospholipid with acetone and solid-liquid ratio of 1:8,60 min ultrasonic time,ultrasonic temperature 45℃,ultrasonic,5 times of low temperature drying for peony phospholipids,phospholipid at a rate of 78.03%,phospholipid content was 97.17%.The content of n-hexane insoluble substance was 0.18%and water content was 0.73%.Determination of 23 fatty acids in peony phospholipid by gas chromatography-mass spectrometry.4 dihydro quercetin and peony phospholipids soluble in ethanol,dry vacuum,the supernatant fluid preparation for peony phospholipids dihydro quercetin compounds,single compound preparation best conditions were optimized as follows:reaction temperature of 40℃,dihydro quercetin and phospholipids ratio of 1:2,dihydro quercetin mass concentration of 10 g/L,the reaction time of 2 h.The final recombination rate of dihydroquercetin was 96%.Fourier transform infrared spectroscopy(FTIR),X-ray diffiaction(XRD)and differential scanning calorimetry(DSC)were used to detect the complex.The infrared spectral structure,crystal structure and melting point peak of the dihydroquercetin phospholipid complex were basically consistent with those of paeoniflorin,indicating that the compound effect of dihydroquercetin and phospholipid was good.The release rate and the cumulative release rate of dihydroquercetin phospholipid compound were significantly lower than that of dihydroquercetin,and the compound had a certain sustained release effect.As a kind of natural and healthy food,the phosphatide of peony has high nutritional value.This paper focuses on the development and utilization of peony phospholipids,in which the complex of dihydroquercetin and peony phospholipids is a major innovation,providing a theoretical basis for the industrial production of peony phospholipids and its application in medicine.
