Styrylpyridine salt was a kind of hemicyanine compound with typical D-?-A structure,moreover,it also had the advantages of excellent optical properties,good stability,good water solubility and easy preparation.It was vital for biological systems and environmental pollution to detect concentration of H2 S and Hg2+.Therefore,this paper constructed three novel H2S/Hg2+ fluorescent probe based on 4-[2-?4-Hydroxy-phenyl?-vinyl]-1-methyl-pyridinium[e]iodide as fluorophore.Chapter 1: In the chapter,the structural characteristics,synthetic routes and applications of hemicyanine were introduced.The research progress of styrylpyridine salt in the field of fluorescent probes was discussed from two aspects: as reaction sites and fluorophores.Based on this,the background,research content and innovation of the paper were putted forward.Chapter 2: A novel ratiometric fluorescent probe DSPBP for H2 S was designed and synthesized,DSPBP adapted 4-[2-?4-Hydroxy-phenyl?-vinyl]-1-methyl-pyridinium[e]iodide as fluorophore and disulfide pyridine group as reaction site.After reacted with H2 S,formed-SH containing intermediate compound,then the ester cyclization proceeded swiftly to yield the fluorophore,it caused obviously fluorescence ratiometric signals changed.DSPBP showed fast response?15 min?,good selectivity,high sensitivity,low detection limit?25.7 n M?and low cytotoxicity.Moreover,DSPBP was successfully applied to the H2 S imaging in living cells and the detection of H2 S in actual water samples.Chapter 3: 4-[2-?4-Hydroxy-phenyl?-vinyl]-1-methyl-pyridinium[e]iodide was used as fluorophore and 2-mercapto-benzyl ester was selected as reaction site to designed a new reactive fluorescent probe MBPVP for Hg2+.The ester hydrolysis reaction of MBPVP was triggered by Hg2+ and accelerated through the strong electron-absorbing effect of fluorophore with positive ion,therefore,the fluorophore was restored and turned on the fluorescence signals.MBPVP showed rapid response time?8 min?,outstanding selectivity and sensitivity for Hg2+ over other metal ions in 100%PBS buffer?p H =7.4?.The linear range was 2-16 ?M and the detection limit was 6.5 n M.Furthermore,the MBPVP was successfully applied to the detection of Hg2+ in actual water samples and in the solid state.Chapter 4: A new outstanding water solubility H2 S fluorescent probe DPPVP was constructed based on thiolysis of dinitrophenyl ether.DPPVP adapted 4-[2-?4-Hydroxy-phenyl?-vinyl]-1-methyl-pyridinium[e]iodide as fluorophore.The high selectivity for H2 S was guarantteed through the significant difference in molecular size and p Ka between H2 S and RSH?biothiol?,when H2 S and RSH coexisted,DPPVP still showed high selectivity for H2 S detection in 100% PBS?p H = 7.4,1 m M CTAB?buffer.DPPVP showed rapid response time,high sensitivity?13.4 n M?and wide linear range?0-40 ?M?.In this paper,three novel H2S/Hg2+ fluorescent probes were based on the reaction of styrylpyridine derivative.They showed good water solubility,excellent biocompatibility,outstanding selectivity and high sensitivity.In addition,they all had good application prospects.
