Study On Purification Of DaDtomycin

Daptomycin,the first cyclo-lipopeptide antibiotics appeared in the market,shows increased indications and good market prospects.The level of biological fermentation has kept stable nowadays;therefore,it becomes top priority to develop a purification process,which is not only different from foreign patents,but also can be applied in industrial production.The main research of this paper is to develop a new purification process of daptomycin by using resin as the main purification material,and high purity daptomycin solution can be prepared by this five-step process including ceramic membrane filtration,macroporous resin purification,anion exchange resin purification,synthetic resin purification,and decolorization by ultrafiltration.Optimized macroporous resin,anion exchange resin and synthetic resin were selected by screening experiments and the main control parameters of the process were investigated and optimized.The final daptomycin product with almost identical quality as commercial daptomycin was prepared by lyophilization process of high purity daptomycin solution after comparing various drying processes.All these studies provide basis for the industrial production of daptomycin and benefit further investigations.
