| Object:This research adopts the hemp seed oil and algae oil mixture is a new kind of plant-based polyunsaturated fatty acids(AH),determine the preparation process of AH soft capsules,and initially investigate its stability.Observe AH on hyperlipidemia model rats blood lipid regulation and anti-inflammatory effects,and explore its possible mechanism of action.Provide theoretical basis and experimental basis for clinical application of polyunsaturated fatty acids in the treatment of hyperlipidemia and even atherosclerosis.Method:1.Optimization of AH soft capsule preparation process The optimization process of soft capsules was determined by orthogonal experiment.2.Initial stability of AH soft capsules The stability of AH soft capsules was initially investigated by influencing factors test,accelerated test and long-term test.3.Study on lipid-lowering,anti-inflammatory,anti-oxidation and mechanism of AH soft capsule After 1 week of adaptive feeding,80 male SD rats were randomly divided into two groups: blank control group(10 rats)and model group (70 rats).SD male rats were fed with high-fat diet for modeling.After successful modeling,the model group rats were divided into 7 groups according to their blood lipid levels: model group,high dose group of AH,medium dose group of AH,low dose group of AH,sea oil group,hemp seed oil group and algal oil group.Body weight,Lee’s index,fat body ratio,blood lipid level,inflammation index and oxidative stress index of rats were measured after 45 days administration,body weight,Lee’s index,fat body ratio,four lipids level(TC,TG,LDL,HDL),Oxidative stress index(SOD,MDA,GSH-Px)and inflammation indicators(CRP,IL-6,TNF-α)were measured in rats.To further detect its lipid-lowering and anti-inflammatory mechanisms,Western-Blot and Real-time PCR were used to detect the expression of genes and proteins related to lipid metabolism(HMGR,PPAR,srebp-1c)and inflammation(AMPK,SIRT1,NF-B,VCAM-1)in rat liver.Results:1.The optimization results of the soft capsule preparation process are as follows: gelatin/glycerin/water ratio is 1:0.5:1,sol temperature is 70 °C;AH soft capsule has good stability,and it is recommended to store in a sealed,dry and cool place.2.AH can significantly reduce the blood lipid level of mixed hyperlipidemia rats,and its mechanism may be related to the expression of lipid metabolism related genes HMGR,PPARα,SREBP-1c.3.AH can significantly alleviate the inflammatory response in mixed hyperlipidemia rats,and the mechanism may be achieved by regulating the expression of AMPK/SIRT1/NF-κB and VCAM-1.4.AH can significantly improve the total antioxidant capacity of mixed hyperlipidemia rats.Conclusion:The preparation process of AH soft capsule prepared in this study is simple and feasible,and the stability is good.AH soft capsule can reduce blood lipid and inflammation levels and enhance antioxidant capacity in mixed hyperlipidemia rats.The mechanism of lipid lowering may be related to the activity levels of HMGR,PPARα and SREBP-1c.The anti-inflammatory mechanism may be related to the activity levels of inflammatory signaling pathways AMPK/SIRT1/NF-κB and VCAM-1.In addition,the lipid-lowering and anti-inflammatory effects of AH in mixed hyperlipidemia model rats were better than those of Hemp and Algal alone,which was comparable to that of seal oil. |
