Abstract/Summary:
Retinal neovascularization disease is recognized as one of fundus diseases,which mainly comprises retinopathy of prematurity,diabetic retinopathy and age-related macular degeneration.This category of diseases seriously imperils the visual health of both infants and adults.Severe retinopathy of prematurity may lead to visual impairment and even blindness in infants and teenagers.At present,the main therapies for retinopathy of prematurity briefly consist mainly of anti-vascular endothelial growth factor treatment and retinal surgery,which have somewhat limitations for infants and teenagers as well as plenty of adverse effects.Thus,it is urgent to develop new therapies of retinopathy of prematurity.Experimental researches imply that GS2005 could reduce the neovascularization of retina and choroid.In order to improve the compliance of infants and teenagers and local drug concentration,this project utilize the cyclodextrin combination,exploiting the stirring method,and fabricating 1.0% GS2005/cyclodextrin suspension eye drops by means of self-assembly.The particles of the suspension eye drops could be retained in conjunctival sac for a relatively long time,with a certain sustained release effect,which could make the drug in the tear fluid saturated,and promote the GS2005 delivery to the fundus.In this article,the pharmaceuticals prescription and preparation conditions were screened by index including inclusion rate,particle size,sedimentation ratio,saturated solubility,and etc.;the inclusion complexes were characterized by X-ray diffraction,Infrared spectroscopy,Transmission electron microscopy,Scanning electron microscopy and Nuclear magnetic resonance hydrogen spectrum,subsequently accepted quality assessment.Finally,GS2005 eye drops were performed pharmacodynamic evaluation in Oxygen-induced retinopathy mouse model.The confirmed pharmaceuticals prescription in this project were as followed: 1.0%(w/v)GS2005,18% γCD,0.7% sodium chloride,0.02% benzalkonium chloride,and 0.1% EDTA.The preparation process was high-temperature sterilization by blank auxiliary solution,GS2005 filtration sterilization,and then stirring in speed at 500 r/min at room temperature for 7d respectively.Finally,the pharmaceuticals were placed in an eye drops bottle.The saturation solubility of the GS2005 clathrate are approximately 60 times higher than that of GS2005.The compounds were characterized by Infrared spectroscopy and X-ray diffraction.The results showed that the inclusion complexes were completely formed,which repeatedly confirmed by Transmission electron microscopy and Scanning electron microscopy.According to Nuclear magnetic resonance hydrogen spectrum test results,it was implied that the inclusion ratio of γCD and GS2005 in the compounds probably was 2:1.GS2005 eye drops quality assessment outcomes are as followed: content and particle size were homogeneous,osmotic pressure,pH value,sedimentation volume ratio was complied to eye drops requirements respectively.In the pharmacodynamic experiment,the results of fundus perfusion fluorescein fundus angiography and optical coherence tomography demonstrated as followed: the formation of retinal neovascularization and vascular tufts in treatment group was significantly reduced,while the growth trend of retinal vessels kept routine.The modified retinopathy scores in treatment group was significantly lower than that of the model group and γCD solution administration group.The heterogeneous interspersion of the inner and outer nuclear layers in retina in treatment group was reduced,which improved severity of retinopathy.The thickness of the retinal optic nerve fiber layer in treatment group was significantly higher than that in model group and γCD solution administration group,which approached that in normal group.The outcomes verified that GS2005 eye drops could effectively treat oxygen-induced retinopathy.
