| Capsules are one of the indispensable products in the pharmaceutical market,which have the advantages of improving the taste of drugs and being easy to use.Animal gelatin is the main raw material of hollow capsules on the market.Animal gelatin has many excellent properties.However,gelatin pollutes air and water greatly in the process of production.In recent years,many plant hollow capsules have appeared on the market.There are many problems in their properties and preparation techniques.For example,Starch capsules are brittle and fragile because they have poor mechanical properties.And starch capsules made from high-strength gel are difficult to disintegrate.In addition,the existing plant hollow capsules have a common problem:capsules are expensive.In order to solve the above problems,a new type of hollow plant capsule is prepared by using celery as the main raw material and potato starch,carrageenan,konjac powder and glycerol as the auxiliary materials.This subject adopts single factor and orthogonal experiment method,taking tensile strength,elongation at break and film-forming properties as performance indicators,using tape casting method to prepare celery fiber plant hollow capsule film,and using hanging film method to prepare celery fiber plant hollow capsules.The mechanical properties and encapsulation properties of capsule films and capsules prepared under different formulations and technological parameters were studied by experiments.Through above works,the ratio of main and auxiliary materials,preparation technological parameters,mechanism of slurry mixing system and the beating law of celery fiber were obtained,and the equipment for celery fiber production was improved.The results are as follows:(1)The optimum preparation formula of celery fiber plant hollow capsule is:celery fiber mass concentration is 3.5%,glycerol mass concentration is 12%,potato starch mass concentration is 18%,carrageenan/konjac powder mass concentration is 0.5%.(2)The optimum preparation parameters of celery fiber plant hollow capsules are as follows:beating degree of celery raw pulp is 35°SR,melting time of slurry is 25 minutes,mixing time of slurry is 6 minutes,mixing temperature of slurry is75?C,water loss rate is 82%,and filtration rate of celery raw pulp is 35%.(3)The mechanism of the mixed system in celery fiber plant hollow capsules:the transmittance test showed that starch,carrageenan,konjac powder and glycerol had good compatibility.SEM showed that good compatibility weakened the phase separation phenomenon caused by celery fiber,and formed a skeleton between fibers,which strengthened the mechanical properties.X-ray diffraction spectra show that a wide diffraction peak and amorphous structure are formed in the mixture system at about 20 degrees.FTIR shows that the absorption peak is weakened near 3300 cm-1,and the mixture system tends to be homogeneous and stable.TG-DSC shows that the capsule film has good thermal stability below 110degrees.(4)Determination of beating regularity of celery fibers:Aiming at the problem of uncontrollable beating degree,the effects of beating time,spacing between flying knife roll and bottom knife groove on beating degree were studied experimentally.The function curve of beating degree with beating time and spacing between flying knife roll and bottom knife groove was fitted by origin software,which provided theoretical support for accurate measurement of beating degree.(5)Improvement design of beater for celery fibre preparation:Aiming at the problems of difficult cleaning and uneven size of beater,it is proposed that the bottom plane of horizontal slurry groove near flying knife roll should be changed to a downward slope to increase the distance between the bottom plane of slurry groove and flying knife roll so as to facilitate cleaning.The uneven fiber blocks can be filtered out by increasing screen mesh to ensure the uniformity of slurry.The celery fiber plant hollow capsule film prepared by this project has good mechanical properties,its tensile strength can reach 13.56 MPa,and its elongation at break can reach 25.35%,which is enough to meet the requirements of capsule filling powder.The prepared celery fiber plant hollow capsule has good cystic property,wall thickness of about 0.2 mm,easy demoulding,bright color,less bubbles,less splitting and thin head,good compactness,moisture content between8.7%and 9%,and drying weight loss between 11.2%and 11.5%.Its viscosity,moisture content,tightness,fragility,length and volume all meet the requirements of international hollow capsules for food and medicine packaging.The results of this study provide a theoretical basis for the preparation of plant hollow capsules with celery fiber as the main raw material,lay a foundation for the large-scale development of new and cheap plant hollow capsules,realize the green and environmental deep processing of celery,and improve farmers’income. |
