| The research object of this experiment was 3,5-2tertiary butyl-4-methyl ether,which was a kind of metabolite that isolate from endophytic etreptomyces rochei of chili.We studied it for making pesticide formulations and biological activity to plant pathogenic fungi,the main research results were as follows:1.Through used single factor,multi-factor orthogonal method and direct emulsification method,we screened for solvent and all kinds of surface active agent,identified 20%3,5-2tertiary butyl-4-methyl emulation in water.eventually formulation was:3,5-2tertiary butyl-4-methyl eher mass fraction of 20%;solvent for xylene,the mass fraction was 20%;emulsifier for(OP-10:polycarboxylate5029:tween-20:rhodia c-630=1:1:3:5),the mass fraction was 20%;thickener for xanthan gum;the mass fraction was 0.02%;antifreeze is ethylene glycol,the mass fraction was 2%;the water mass fraction was 38%.The preparation of water eventually formulation indicators were qualified,after stored at room temperature,heat and cold,it was no oil analysis and layered phenomenon occurs,the prepared formulation viscosity was small,the dumping of sex was better also,it contributed to storaged and packaged2.Through used single factor,multi-factor orthogonal method and direct formula repeated trials,we screened for carriers and all kinds of surface,identified 20%3,5-2tertiary butyl-4-methyl wettable powder eventually formulation was:acephate technical mass fraction of 20%;wetting agent lauryl sodium sulfate,the mass fraction was 3%;wetting agent silok-7110,the mass fraction was 4%;wetting agent NP-400,the mass fraction was 2%;dispersant calcium lignosulphonate mass fraction was 4%;dispersant sodium lignosulphonate mass fraction of 4%;dispersant carboxymethylcellulose mass fraction of 10%;dispersant polyvinylpyrrolidone mass fraction was 8%;finally,diatomaceous earth make up 100%.Accorded to the national standard of wettable powder,the suspension rate of the agent,wetting time,fineness,pH,hot and cold storaged were up to standard.3.Through growth rate method,we measured 20%3,5-2tertiary butyl-4-methyl emulation in water,20%2,6-Di-tert-butyl-4-methyiphenol emulation in water,two kinds of dosage forms of plant pathogenic fungi hyphae growth inhibition,and two kinds of water emulation mixed matched to Watelmelon Fusarium oxysporum,Botrytis cinerea Pers.,F.graminearum virulence determination,single emulsion in water toxicity measurement results show that:There was a certain inhibition effect of two formulations against the pathogenic fungi,the effect of inhibiting mycelia growth which was most obvious against F.graminearum,the median effective concentration(EC50)was 2.64 mg·L-1 and 22.07 mg·L-1,Mixed joint toxicity results showed that:When the two kinds of water emulsion mixed ratio of 1:9,there had highest synergistic ration of three kinds of pathogenic bacteria,respectively was 3.96、5.00、4.47。4.The mechanism of bacteriostasis research showed that:treated by the 20%3,5-2 tertiary butyl-4-methyl emulation in water,pathogen hyphae distorted out of shape,some happened ruptures.Ultraviolet spectrophotometry detection showed that the liquid after treatment,the nucleic acid,protein levels in it were significantly elevated,it showed that the bacteria cell membrane permeability changeed and could speculate that the actions sites may be on the cell membranes of pathogenic fungi.5.The test results of protect enzyme activity showed that after spray processing which used the 20%3,5-2 tertiary butyl-4-methyl emulation in water,four kinds of protective enzyme in stems and leaves in general present the form of growing,after one week,they were significantly higher than the control group.It showed that those emulsion had the ability to improve plant resistance to disease.6.20%3,5-2 tertiary butyl-4-methyl emulation in water for tomato seed germination and seedling growth measurement results showed that:the water emulsion had a certain role in promoting to seed germination,and dry weight,root length in low concentration of 6.25~25 mg·L-1,but with the concentration rised,it had inhibitory effect.Equally water emulsion in low concentration of 6.25~25 mg·L-1 can contribute to the growth of tomato seedlings,but with the increased of concentration,it played an inhibitory effect on the tomato root length,fresh weight of aerial part and underground part fresh weight7.The determination of 20%3,5-2 tertiary butyl-4-methyl emulation in water control effect on A solani by using pot method,the results showed that the water emulsion had a role in the prevention and treatment for A.solani,when concentration was 400 mg·L-1,water emulsion for A.solani prevention effect is the best,within 24 hA.solani was inoculated and after 5 d the defensive efficiency reached 77.27%,within 120 h A.solani was inoculated and after 5 d defensive effect was 69.04%.The prevention effect of 20%3,5-2 tertiary butyl-4-methyl emulation in water on the surface of the leaf was better than treatment effect.With time expanded and concentration reduced,the control effect gradually begin to fall. |
