In functional foods,the development and utilization of natural products are increasingly being regarded as an advantageous item in the natural products industry.This kind of functional food represented by natural products has a clear,specific and reliable health function.In this experiment,natural products and corn starch were used as substrates to form water-soluble products under the catalysis of high temperature transglycosylase.The yield of glycosylated products reached a certain limit.By further changing the gene base of its wild bacteria,the highly active mutant enzyme was screened,thus improving the glycosylation activity of the enzyme and increasing the yield of the glycosylated products.In this experiment,the effect of non starch polysaccharide colloid on the rheological properties of starch was studied,and the optimum compound system was obtained.1h was prepared by reaction of sophorae,corn starch,pH6.5 phosphate buffer solution and high temperature resistant enzyme at 85 C.Glycosylation of fructus sophorae and cornstarch occurred.For different concentrations of substrate,the amount of enzyme,buffer and DMSO also changed.In order to increase the solubility of sophoroside and increase the proportion of DMSO in the reaction system,the maximum solubility of sophoroside reached 28.3mg/mL.With the increase of substrate concentration,the yield of new product of sophoractone also showed an upward trend.When the volume ratio of DMSO reached 40%,the yield tended to be 74.2%.The products were treated to remove the reducing sugar and DMSO.The results showed that the yield of sophoroside was reduced and the product was lost in the treatment process,and the yield was 54%.In the HPLC test,the concentration of sophoroside in the reactant was 32mg/mL,the solubility of sophoroside was 7.88 mg/mL,the solubility of sophoroside reactant increased and the water solubility increased.The scavenging rate of DPPH radical of the same concentration of sophoroside and its products is different.The scavenging effect of sophoroside products is improved.After analysis,the free radical scavenging rate of the solution was significantly different when the concentration of solution was 2.1mg/mL and 2.8mg/mL.The kinetic parameters of enzyme were obtained from amylase and amylopectin solution by using mutagenesis enzyme and wild enzyme respectively.Among the above substrates,the Km value of amylopectin(mutagenesis)is larger than that of amylose(wild enzyme),and the Km value of amylose(mutant enzyme)14.96mg/mL is larger than that of amylose(mutant enzyme)Km value 6.57mg/mL,which indicates that the affinity of wild enzyme to substrate is stronger than that of mutant enzyme,but it is better than that of mutant enzyme,but the Km value of amylose(mutant enzyme)is stronger than that of mutant enzyme,but it is better than that of mutant enzyme.The Kcat value of the mutant enzyme is far greater than the Kcat value of the wild enzyme,indicating that the number of molecules that each enzyme can be converted to the substrate is stronger than that of the wild enzyme within the unit time of the mutant enzyme.The Kcat/Km of the mutant enzyme is larger than the wild enzyme,and the catalytic efficiency of the enzyme is higher than that of the wild enzyme.The enzyme can improve the utilization ratio of corn starch as a sugar donor.In the transglycosylation of mutant enzyme and wild enzyme with genistein,the yield of the new product from wild enzyme was 20%,and the yield of the mutant product was 22%.In order to study the effect of non starch polysaccharide colloids on the rheological properties of starch,the viscoelasticity of 5 different mixtures of robinia pseudoacacia gum/corn starch(0:1.2,0.1:1.1,0.2:1.0,0.9:0.3,0.4:0.8,mass ratio,g/g)was measured.The Power-Law model was used to regress the static rheology test data.The determination coefficient R~2 was above 0.99,and the fluid index was n<1.It showed that the corn starch and the mixed system belong to pseudoplastic fluid.With the improvement of the mass ratio of the mixture of robinia bean gum/corn starch,the consistency coefficient of the mixture system increased significantly,and the fluid index was significant.The number is reduced and the pseudoplastic is enhanced.When the robinia bean gum/corn starch mass ratio is less than 0.3:0.9g/g,the consistency coefficient is no longer significant.The dynamic rheology test showed that when the mass ratio of locust bean gum to corn starch was greater than 0.3:0.9g/g,the elasticity was higher and the viscosity was moderate.When the mass ratio of locust bean gum to corn starch is 0.4:0.8g/g,the viscosity is the largest and the elasticity is the smallest.The mixed system has better synergistic thickening effect.
