With the rapid development of domestic breeding industry,people pay attention to the vitamin supplement of animals to improve their immunity.Due to the deficiencies of traditional multivitamin products,new vitamin product with high efficiency,stability and safety is urgently demanded in the breeding market.In recent years,multivitamin nanoemulsion as a new vitamin supplement product has become the first choice for aquaculture industry with its unique advantages.However,there are still some problems such as low content of vitamin B2(VB2)and vitamin B9(VB9)and poor stability.In this study,supercritical fluid technologies such as supercritical anstisolvent-(SAS)and particle formation from gas-saturated solution-(PGSS)and SAS-assisted inclusion were proposed to solubilize VB2 and VB9 in order to increase the solubility to 100 mg/100mL and 80 mg/100mL,separately,in water.Then,a multivitamin formulation was synthesized by using the micro emulsification technology;the stability was tested.The two different supercritical fluid-assisted technologies(SAS,PGSS)for preparing microparticles were firstly employed to deal with VB2,and results showed that the solubility of VB2 in water was increased from 12.00 mg/100mL to 28.02 mg/100mL by using SAS,while it increased to 20.37 mg/100mL by using PGSSS.Then the traditional mixing method and SAS-assisted inclusion method was applied to VB2 with different vitamin carriers,and results showed that the solubility of VB2 in water was increased to 51.37 mg/100mL by the traditional mixing method,while it increased to 140.28 mg/100mL when using 2,6 dimethyl-β-cyclodextrin as the vitamin carrier.It can be concluded that 2,6 dimethyl-β-cyclodextrin was the best vitamin carrier,and the SAS-assisted inclusion is the best way for enhancing the solubility of VB2 in water.Consequently,the SAS-assisted inclusion was also applied to VB9,and results showed that the solubility of VB9 was increased from 0.16 mg/100mL to 82.33 mg/100mL after the treatment.The micro emulsion technology was employed to prepare a multivitamin formulation by using the fat-soluble vitamins A,D,E,K,the water-soluble vitamin B1,B6,B12,biotin,pantothenic acid,nicotinamide,and the as-prepared VB2 and VB9 from the SAS-assisted inclusion method.,The formulation showed good stability when 0.3%stabilizer and 0.3%antioxidant was additionally mixed,no precipitate and color change could be observed in 120 days.
