Peptic ulcer is a common and multiple chronic disease of the digestive system,mainly including gastric ulcer and duodenal ulcer,which about 10%people in the crowd suffer from.Drugs which are currently used to treat peptic ulcer mainly include antacids,H2 receptor 'antagonists,proton pump inhibitors and mucosal protective agents.However,adverse reactions caused by antacids,H2 receptor antagonists and proton pump inhibitors have often been reported.Based on the above status of peptic ulcer drugs,the preparation process and performance evaluation of calcium hydroxide antacid loaded on montmorillonite,which is a new antacid,have been studied in this paper.Calcium hydroxide antacid not only greatly reduces the incidence of adverse reactions,but also exerts the protective effect on mucosa.Considering both in-vitro acid resistance and solubility of acid resistant active ingredient Ca?OH?2,the optimum process conditions are as follows:under conditions of Ca?OH?2 content of 37%,reaction temperature of 75? and feeding time of 4 h,lime milk was uniformly dropwise added into acid-activated montmorillonite.When Span 60 was used as a surface modifier,with the optimum dosage of 2 wt.%,time up to pH3 of imitative gastric juice was as short as 3 min,and time between pH3-5 of imitative gastric juice reached 49 min,in the process of in-vitro acid resistance.As stearic acid was used as a new surface modifier,process simplification and improvement not only reduced the amount of surface modifier,but also made calcium hydroxide perform more excellent in the in-vitro acid resistance.Under condition of stearic acid optimum dosage of 0.5 wt.%,time up to pH3 of imitative gastric juice was less than 3 min,and time between pH3-5 of imitative gastric juice was as long as 57 min.Acute toxicity test and long term toxicity test were studied on mice with calcium hydroxide antacid,so as to identify whether calcium hydroxide antacid has any side effects and get basic data for clinical safe medication.In acute toxicity test,death,toxic effects,visceral organs disease and damage have not occurred in the mice.As the dosage of calcium hydroxide antacid was as high as 8000 mg·kg-1,according to globally harmonized classification system,calcium hydroxide antacid belonged to toxicity of unclassified for 8000 mg-kg-1>5000 mg·kg-1.And according to EU toxicity grading standards,calcium hydroxide antacid also belonged to toxicity of unclassified for 8000 mg·kg-1>2000 mg·kg-1.In long term toxicity test,daily dosage of calcium hydroxide antacid was up to 500 mg·kg-1 and the antacid was continuously delivered for 15 days.Death and toxic effects have not occurred in the mice,and the mice lived in good condition as before.In addition,visceral organs showed healthy and normal,without disease and damage.Therefore,two types of toxicity test showed that calcium hydroxide antacid was a safe drug.Calcium hydroxide antacid performs both acid resistant property and gastric mucosal protective effect.In gastric mucosa protection experiment,the effect of calcium hydroxide antacid on gastric mucosal protective effect was studied when the dosage of surface modifier was different.Results showed that both the modified and unmodified calcium hydroxide antacid performed gastric mucosal protective effect,and calcium hydroxide antacid performed the best gastric mucosal protective effect when the dosage of stearic acid was 0.5 wt.%.The effect of commercially available hydrotalcite antacid and different dosage of calcium hydroxide antacid on gastric mucosal protective effect was studied as well.Results showed that both commercially available hydrotalcite antacid and calcium hydroxide antacid performed gastric mucosal protective effect.Besides,low-does calcium hydroxide antacid was a little worse than commercially available hydrotalcite antacid in gastric mucosal protective effect,and middle-does as well as high-dose calcium hydroxide antacid were similar to commercially available hydrotalcite antacid in gastric mucosal protective effect.Calcium hydroxide,supported on montmorillonite,was first modified for internal medicine.The carrier montmorillonite has improved the dispersion of Ca?OH2?2,so the solubility of Ca?OH?2 significantly reduced and a protective barrier was formed.Therefore,calcium hydroxide antacid performs excellent anti peptic ulcer effect.In addition,calcium hydroxide antacid exhibits potential application in calcium supplement and acid constitution improvement.
