Phellinus baumii has role of anticancer,antioxidant and antimicrobial,which is a valuable Chinese herbal medicine.Phellinus baumii is studied mostly in South Korea,Japan,Europe and United States at present.The polysaccharides and flavonoids are studied mainly,but the pigment obtained from Phellinus baumii has rarely been reported.Natural edible pigment has a double role of colorants and antioxidants,it has not only color-enriching,non-toxic and non-side effect,also has effect of antioxidant,anti-tumor and so on.which has been widely used in the new food additive.So it is meaningful to study the pigment obtained from Phellinus baumii.The strains of Sanghuang were identified,the best conditions of extraction,purification of pigment,the properties and components of pigment were studied in this paper.The Sanghuang was Phellinus baumii through culture characteristics firstly,observation under electron microscope then and molecular detection identification.It was sieved firstly and through 10 mesh then.The crude pigment was extracted by ethanol.The optimal extraction conditions of pigment of Phellinus baumii were got through the single factor experiment and response surface design.The best extraction conditions were following:liquid-solid ratio was 1:36 g/mL,ethanol concentration was 62%,extraction temperature was 74?,extraction time was 4.0 h,the absorbance of pigment was 26.25.By comparing adsorption rate and desorption rate of DM301,H103,D101,AB-8to Phellinus baumii pigment,the results showed that the DM301 resin had the best purification effect on pigment.The optimal purification conditions of macroporous resin DM301 from the static test and dynamic test were following:The volume of pigment was 60 mL,the best adsorption pH were 2,the time was 60 min,the best desorption pH was 3,ethanol concentration was 70%,the elution flow rate was 2BV/h,the ethanol volume was 240 mL.The color value was 71.68 after purification,which was about 9 times before purification of pigment.Pigment has good tolerance to acid,even in 2 mol/L citric acid and tartaric acid,the absorbance of pigment extracted from Phellinus baumii was unchanged after 1days,the absorbance remained about 0.48,the retention rate was 100%.Pigment had strong tolerance of heat,even was heated at 100?for 5 h,the retention rate of absorbance was still more than 90%.It was more stable in the small polarity material.The retention rate of pigment can still reach to 75%after 15 days although in outdoor.But most of mental icons had great influence on pigment,the most influence of mental icons was Al3+and Fe3+.When the concentration of Al3+and Fe3+were 50?mol/L,the retention rate of pigment was less than 20%.Only Na+had no effect on the pigment.Therefore,the pigment should avoid mental ware during transport and storage.The studies on antioxidation of the pigment showed that pigment has strong antioxidant activity,the reducing capacity of pigment was slightly higher than that of Vc,the hydroxyl radical?·OH?scavenging ability and DPPH·scavenging capacity was much higher than that of Vc,when the concentration of pigment was 0.4 mg/mL,the hydroxyl radical?·OH?scavenging ability reached 100%.,but VC was 50%.When the concentration of pigment was 0.025 mg/mL,the DPPH·scavenging capacity was85%,which was 4 time of VC.The superoxide anion(O2-·)scavenging capacity of pigment was less than Vc.When the concentration of pigment was 50?g/mL,the superoxide anion(O2-·)scavenging capacity was about 40%,when the concentration of pigment was 100?g/mL,the superoxide anion(O2-·)scavenging capacity can reach about 60%.In addition,the pigment obtained from Phellinus baumii had protective effect on DNA,it can make the fluorescence intensity weaken.Through the study of these properties showed that the pigment obtained from Phellinus baumii not only can be used as food and medicine colorant to improve color and appetite,but also can be as an antioxidants and DNA protective agent to effectively remove the harmful free radicals of body and protect DNA.The pigment was identified by IR and LC-MS.The results of IR show that the pigment purified by macroporous resin in the 420 nm contains a lot of material and have many functional groups and structure,it need purify further.After LC-MS analysis,it showed that 16 kinds of main compounds in the pigment obtained from Phellinus baumii were identified,they account for 86%.but they all were not found the corresponding components.The main material of pigment obtained from Phellinus baumii contains carbon,hydrogen,oxygen and nitrogen,it still need more and deep study.
