Tylosin is a Streptomyces fradiae emergence of 16 yuan macrolide veterinary efficient non-existent antibacterial and promoting agent,at home and abroad have a common market prospects,the market demand is huge.At present,in the production process,there are low titer of bacteria,fermented raw materials,fish meal,soybean oil and other high prices,and even higher than the production cost of the sale price of the situation.In order to solve the above problems,the strain of Streptomyces fradiae TP116 was mutagenized by triple recombination mutagenesis to obtain high-yield strain,and then the culture conditions of the resistant mutant and the ratio of the raw material of the main fermentation medium were Optimize the improvement,in order to give full play to high-yield strains of production capacity,improve the level of industrial fermentation tylosin,reduce production costs and enhance product market competitiveness.This study leads to the following main results:(1)After microwave irradiation for 30 s and UV irradiation for 60 s and lithium chloride,the strain was then screened by adding tylosin powder to the traditional screening method and the stability experiment was carried out to obtain strain TPF116-27,titer Reached 10656u/ml,which was 14.3% higher than that of the original strain,and was used as a subsequent shake flask fermentation strain.(2)The optimum initial pH value of the seed culture medium was 7.0,the culture temperature was 30?,the age was 46 h,and the filling volume was 45 ml.Through the optimization of seed culture conditions,the quality of the seeds was greatly improved,and the cultivation period of the seeds was shortened,which laid a foundation for the subsequent production of tylosin.(3)The main medium ratio of tylosin fermentation was determined by single factor experiment and orthogonal experiment.0.15% hydrochloric acid betaine was added at the beginning of fermentation for 4 hours,and then the concentration of cobalt was 0.20% Sodium acetate,2.0% fishmeal,rapeseed oil 6.0%.TPF116-27 was obtained by bacterial mutagenesis,and the fermentation conditions were optimized.The titer of tylosin was 11876u/ml,which was 27% higher than that of the original strain.These results provide a reference for enterprises to further improve and improve the industrial fermentation level of tylosin,improve production efficiency and reduce production costs.
