Preparation Of Xingjiuhugan Chewable Tablets

On the basis of ethanol metabolic pathways and mechanism of drug action,an antialcoholism and hepatoprotecion prescription was obtained in the present paper.The purpose of this project was to make the prescription to be chewable tablets with good taste which was more convenient for patients to take.The main contents were as follows:1)An mice drunk experiment model was built to investigate the antialcohol effect of prescription 1~5,using drunkenness and sober time as index.Rusults indicate that prescription 1 has the good antialcohol activity.2)In this study,a HS-GC procedure for determination of ethanol and acetaldehyde concentration on rat blood was established.The effect of XJHG(prescription 1)on the pharmacokinetics of ethanol in SD rats following oral administration was investigated.The results showed that the treatment of rats with XJHU decreased AUC0-12 h of ethanol by 66.06% and Cmax of ethanol by 51.60%.CL/F of the treated group was 9.17 multiple higher than that of controlled group(P<0.05).3)The effects of XJUG against alcohol induced liver disease in mice were investigated.The results showed that high and middle dosage resulted in a decrease in serum ALT and AST level.Besides,co-treatment of XJHG also enhanced GSH contents,but decrease liver index,MDA and TG contents in liver,weaken liver steatosis(P<0.05 and P<0.01).4)Single factor test and orthogonal test were applied to screen the formulation and preparation technology of XJHG chewable tablets.And the optimized formulation was as follows: extract dosage 58.3%,mannitol 28.7%,lactose 11.4%,aspartame 0.5%,citric acid 0.1%,appropriate 70% ethanol,magnesium stearate 1.0%.The critical relative humidity of granule was 73%.5)Content determination of XJHG chewable tablets was investigated.We adopted UV to determine the content of polysaccharides and total flavonids in XJHG chewable tablets.Puerarin was determined by HPLC.The methods were simple,accurate and reproducible.
