Cordyceps militaris,also known as north Chinese caterpillar fungus.The active components of cordyceps militaris are mainly adenosine derivatives,polysaccharides,cordyceps acid,ergosterol and manniyol,ect.Thesee componets have been reported to exhibit various activities including immunity boosting,anti-tumor,anti-viral,anti-inflammatory,anti-oxidation,and anti-aging.The development of Cordyceps militaris-based functional drink may have good market prospects,since they take fully adventage of all the active ingredients of Cordyceps militaris,as well as other nutritional ingredients.In this thesis,Cordyceps militaris active ingredient extraction technology and the related functional drink production process were optimized,quality control methods were established,and the evaluation of the functional drink were also performed.1、Single factor analysis method was used to investigate the impact of solid-liquid ratio,extraction temperature,extraction time,and extraction times on cordycepin and adenosine extraction rate.The optimizedextraction peocesses were as follows: extraction temperature: 58.0 ? C,extraction times: 3.0 times,extraction time: 3.4 h,liquid ratio: 36.0 mL/g.2、A HPLC method was established for simultaneous determination of cordycepin and adenosine.The method was established with good system suitability,accuracy,precision,reproducibility,recovery and liner relationship in predetermind range.It can be used for content determination of cordycepin and adenosine.3、Preparation methods of functional drink of cordyceps militaris were studied.DTH determine that 1.0g cordyceps militaris extract was added250 mL functional drink.A HPLC method was established for simultaneous determination of cordycepin,adenosine,nicotinamide and caffeine.The method was established with good system suitability,accuracy,precision,reproducibility,recovery and liner relationship in predetermind range.It can be used for content determination of cordycepin,adenosine,nicotinamide,and caffeine.4、The functional drink was orally administered to mice at three does(2.08 mL/kg·bw,4.16 mL/kg·bw,and 12.5 mL/kg·bw)for 35 days.The result showed that the functional drink can boost the immunity.In addition,the mice droppings were collected and analyzed for the intestinal microflora variation by metagenomic technology.The result showed that the functional drink can change the intestinal microflora of mice.It was concluded that the functional drink can regulate intestinal microflora anddeserve further study for verification.
