Cordyceps militaris as a kind of precious edible-medicinal fungi,It has been highlyconcerned by scientists. The polysaccharide from Cordyceps miLitaries is one of the mostimportant components. Recently, the immune regulation, anti-tumor and antioxidant activity ofthe polysaccharide have been perceived by peopLe. Ultrasound-associated extractingpolysaccharides from Cordyceps militaris, separating and purificating polysaccharides and theactivity are the main research contents in this thesis. And the resuLts are as follows:1. With the ultrasonic frequency, the power, the time, the temperature, the ratio of materialand water and the powder size of Cordyceps militaris, which are as the base. Then, the power,the time, the temperature and the ratio of material and water are conducted by the responseanalysis, the resuLts show that the optimum conditions about the extracting polysaccharides arethe220.258w of the power, the50.244min of the time, the70.564℃of the temperature and the30.998of the ratio of material and water, and the optimum extraction ratio can be7.174%.2. After getting polysaccharide from Cordyceps militaries, isolation and purificationpolysaccharides were studied. The successive steps were concentration, Sevag method toeliminate protein, AB-8macroreticular resin to wipe out pigment and dialyzing to removemicromolecule. In order to get pure polysaccharide, DEAE-52Cellulose anion as infilling begancolumn chromatography purification, getting two kinds of components, which were named F1and F2. The content of F2was higher, which was the main content of the Cordyceps miLitarispoLysaccharide and also was the active polysaccharide. These two contents were furtherpurification by sephadex G-100, which get a single peck. This result indicates F1and F2are solecontent.3. The anti-tumor effect of Cordyceps militaries polysaccharide in vitro was studied. Theresult shows that this kind polysaccharide can control the growth of cancer cell. When theconcentration of the F2was2500ug/mL, the ratio of inhibition to melanoma cell A-375can get to31.65%. When the concentration of the F2was2000ug/mL, the ratio of inhibition to cancer ofLiver cell HepG2can get to27.9%.4. The antioxidant effect of Cordyceps militaries polysaccharide in vitro was studiedthrough clearing hydroxyl free radical test, clearing superoxide test and the determination of thetotal reduction ability test. The results show when the concentration of Cordyceps militariespolysaccharide was25mg/mL, the ratio of inhibition to hydroxyl free radical can get to70%andthe ratio of inhibition to superoxide can get to75%, when the concentration of Cordycepsmilitaries polysaccharide was20mg/mL, the absorbance in700nm was1.068, which shows this kind of polysaccharide has good reducing capacity. The antioxidant effect and the concentrationof Cordyceps militaries polysaccharide were concentration-response dependence.5. Establishing mouse models of D-galactose induced aging, which were studied anti-agingtest. These mouse were forced different concentrations of Cordyceps militaries polysaccharideinto stomach everyday for42days, then take off the cervical of these mouse, and study theirimmune organs, the SOD, GSH-PX and MDA of liver and serum.With analysis of variance todeal with the results, the optimal anti-aging effect was medial-dose group(400mgpolysaccharide/kg weight). |
