| Objective:Evening primrose(Oenothera biennis.L)is a plant of perennial Onagraceae herb,which is mainly produced in the easthern mountainous of northeast area of China.Evening primrose oil is extracted from the mature seeds,.The main active ingredient is y-linolenic acid but unstable in itself,To overcome this nature y-linolenic acid,evening primrose oil to prepare a flexible nano-liposomes.In this study,The evening primrose oil encapsulated in liposomes to optimize the flexible nano-preparation process,and evaluated the quality and pharmacodynamics observed.Methods:Prepared evening primrose oil Flexible nano-liposomes with using the solvent evaporation,As a study of factors by evaluation of evening primrose oil Flexible nano-liposomes’s encapsulation efficiency,by evening primrose oil and lecithin quality ratio、lecithin and sodium cholate quality ratio、cholate and cholesterol mass ratio.Using the L9(34)orthogonal design to optimize preparation of evening primrose oil flexible nano-liposome.The quality appraisal and stability of vening primrose oil flexible nano-liposomes by gas chromatography,laser particle size analyzer,TEM detection methods.Using TP-6 transdermal diffusion to instrument percutaneous,research transdermal rate of evening primrose oil flexible nano-liposome.Through the rabbit skin irritation test on evening primrose oil Flexible nano-liposomes for safety inspection and evaluation.The mice using croton oil mixture make ear swelling to study of evening primrose oil Flexible nano-liposomes anti-inflammatory effect.Using a hot plate method and mice writhing method to research calmative effect of evening primrose oil flexible nano-liposome.Results:The final formulation of evening primrose oil flexible nano-liposomes is evening primrose oil 300 mg,evening primrose oil and lecithin mass ratio of 1:10,lecithin and sodium cho late mass ratio of 10:1,Cholate and cholesterol mass ratio of 1:3,pH 7.0 of phosphate buffer solution,the average encapsulation efficiency was 90.31%.Evening primrose oil Flexible nano-liposomes as a milky white homogeneous suspension body and good fluidity.The average particle diameter of 125 nm,Zeta potential is+52.15 nm.In transmission electron microscope,the particles are spherical,rounded appearance,good dispersion.Determination of y-linolenic acid of evening primrose oil flexible nano-liposome content by gas chromatography,γ-linolenic acid methyl ester in 0.02064~0.14265 mg/mL range have a good linear relationship,This method recovery RSD is 0.83~1.45%.Three batches of evening primrose oil flexible nano-liposome average γ-linolenic acid content of 0.732%.Stability tests showed,flexible nano-liposomes at 4℃ storage conditions are more stable.Transdermal permeation experiments,evening primrose oil flexible nano-liposomes better than the average evening primrose oil for transdermal performance and high skin cumulants.Pharmacodynamics showed,evening primrose oil Flexible nano-liposomes with anti-inflammatory and analgesic effects,and high-dose slightly better than the positive control group Voltaren ointment.Conclusion:Evening primrose oil flexible nano-liposome preparation process is simple,high encapsulation efficiency and good stability,quality to meet the requirements,characteristics of physical and chemical properties are stability.In vitro transdermal experiments showed evening primrose oil flexible nano-liposome better than evening primrose oil in penetration and promoting.evening prinuose oil Flexible nano-liposomes no obvious skin irritation in rabbits.Evening primrose oil flexible nano-liposomes have significant analgesic and anti-inflammatory effects... |
