~1H NMR-Based Metabonomic Study On Acute Biological Effects Of Haematitum-Treated Rats

Abstract/Summary:
Mineral medicine is a natural mineral formed by geological processes.In a certain range,its chemical composition is stable,has a very important medical value.Haematitum belongs to contain iron mineral medicine(Fe2O3),its iron content more than 45%,is good supplement of iron,can be used for the treatment of iron deficiency anemia,regulating endocrine balance etc.In recent years,Haematitum has been the subject of extensive studies on processing methods and clinical application,while less researched in its endogenous metabolism.In this paper,we use ~1H NMR-based metabonomic analysis coupled with multivariate statistical analysis(principal component analysis and partial least squares discriminant analysis)to analyze the metabolic profiles of the collected urine samples and serum samples from Haematitum treated rats,observe liver tissue and kidney tissue pathology,and study the changes of the serum biochemical indexs from the collected serum samples.The same significant treatment related changes in all dose treated group were observed for the levels of Tuarine,Creatinine,α-Ketoglutarate,Lactate,Acetate,Choline,Glucose,Low density lipoprotein and very Low density lipoprotein,which can be used as potential features of haematitum.The different changes in the low dose treated group were observed for the levels of Choline,Isobutyrate,β-Hydroxybutyrate,Lipid.The different changes in the medium dose treated group were observed for the levels of Succinate,Dimethylglycine,N-Acetyl glycoprotein1,N-Acetyl glycoprotein2.The different changes in the high dose treated group were observed for the levels of Citrate,Succinate,β-Hydroxybutyrate,Creatine.A trend of recovery in connection with dose levels was observed overtime.The variation of biochemical index from all dose treated group included Lactate dehydrogenase,Alanine aminotransferase,Aspartate transaminase,Alkaline phosphatase,Cholinesterase,Triglycerides,Total cholesterol.And in the high dose treated group,we observed the levels of Blood urea nitrogen changed.Such biochemical changes indicated that Haematitum treatment at all dose of 2,5,10g/kg body weight affected the Krebs cycle and glucose metabolism and energy metabolism in rats.Haematitum treatment at the medium dose affected dimethylglycine metabolism.These changes may be attributed to the disturbances of liver function in all dose treated group.And these changes also may be attributed to the disturbances of hepatic function in high dose treated group.From the liver tissue and kidney tissue pathology picture,we didn’t find any changes of cells’ shape of the liver tissue and kidney tissue.This may indicated that the effect of Haematitum on liver function and hepatic function were not very serious.From the above,~1H NMR-based metabonomic can be effectively applied to the study of Haematitum and to predict possible potential characteristic metabolites of Haematitum.
