Research On Pharmacy Part Of Hua Bian Mixture In Pre-clinical

Hua Bian mixture is a clinical prescription of the emergency department of the affiliated Hospital of Chengdu university of Traditional Chinese Medicine.It is composed of 10 drugs.After years of clinical experience,it is proved that this prescription has the efficacy of clearing heat and detoxifying,torrey and evil,and is used to treat acute tonsillitis and pharyngitis.In this study,It is based on the traditional Chinese medicine theory as the guiding principle,and use the modern technology technology for the pre clinical pharmacy part,including preparation process,molding technology,quality standards and stability.According to the prescription function and physicochemical propert of the effective ingredients,the preparation process was prepared.The preparation technology includes three aspects:extraction,concentration and purification.In the extraction process,the aromatic water was extracted by steam distillation.By determining the content of the volatile oil,the extraction time was determined to be 6h.In consideration of the total volume requirement of the preparation,the aromatic perfume was distilled into medicinal herbs:aromatic water was about1:1.The dregs and the other eight flavors were extracted,With composite scores of extraction amounts of chlorogenic acid,baicalin,phillyrin,arctiin and dry paste yield as the comprehensive evaluation index,orthogonal test of L₉?3⁴?was adopted to investigate effects of the amount of water,decocting time and times on extraction process.The above indicators were comprehensively scored through information entropy theory.Water extraction technology was as follows:added 8-foLd water,each time for 1 hour.The results showed that except for arctium,the transfer rates of the above three components reached over 70%.In the study of concentration and purification process,due to the instability of chlorogenic acid,it is determined as decompression concentration and centrifugal purification.The centrifugal time,speed and concentration of Liquid were also investigated.Finally,it was determined that the ratio of the decompression concentration to the Liquid ratio was 1:4,and the centrifugation?speed:6000r/min,time:30min?and the concentration to the Liquid ratio of 1:1.In the study of molding process,Mix the concentrated liquid with the aromatic water,Determine the amount of prescription,and Factors affecting the stability of the stabilizer?light,temperature?,packaging materials,pH range,type and amount of additives were investigated.Finally determine the molding process is:Select potassium sorbate as a preservative,the amount of 0.1%,stevia is a flavoring agent,the amount of 0.3%,adjust the pH value of 5.0-6.0,polyethylene brown plastic bottle filling,sealed,indoor temperature and avoid light.In the study of quality standard,tests are conducted on both the TLC identification and the content determination.The identification method of fritillaria thunbergii,trichosanthin and forsythia forsythia was simple,reproducible and special,the income standard,the negative of fructus arctii was disturbed,At the same time,a multi wavelength switching HPLC method was established for simultaneous determination of chlorogenic acid,forsythin,baicalin and arctium,and the method was studied.The results are all in accordance with the requirements.According to the content determination result of the sample,the content limit is stipulated,the content of the chlorogenic acid should not be less than 2.9 mg·mL^-1,and the content of baicalin should not be less than 6.5 mg·mL^-1,and the content of Forsythia glycoside should not be less than 0.4mg·mL^-1,and the content of arctin should not be less than 1.2 mg·mL^-1.In the study of stability,the 6 month accelerated test and 12 month Long term test were carried out.The results were all in line with the requirements.It showed that the preparation process was reasonable,the quality controlled and the stability was good.
