As we all know,traditional Chinese medicine with anti-cancer activity generally has a multi-target,multi-link,multi-effect of anti-tumor,besides,it has low side effects,are not easy to produce resistance and not genotoxic.Therefore,the anticancer activity of Chinese medicine has become the current research hot spots.The Stellera chamaejasme in our country has a long history of medication and the main drug site is the root.The modern pharmacological studies have shown that its extract has a strong anti-tumor and anti-virus activity.The research about chemical composition of its extract and correlation with the anti-tumor activity has a very important significance.In this work,the chemical composition of different polar solvent extracts from root of Stellera chamaejasme L.were analyzed by modern instrumental analysis technique.And the correlation between its chemical composition and activity was discussed preliminarily with the in vitro activity test.The main research contents and results are as follows:1.The establishment of chemical composition analysis about the hydrocarbon extract which came from the root of Stellera chamaejasme L.The conditions for the separation and detection of hydrocarbon extracts from root of Stellera chamaejasme were optimized systematically.It established the enrich method of solvent extraction,HP-5 column separation and gas chromatography-mass spectrometry?GC-MS?analysis of the roots of Stellera chamaejasme L.hydrocarbon analysis of the method.55 components were isolated and 44 structures were identified,accounting for 97.73%of the total area.Among them,there are 11 hydrocarbon substances,10 aromatic substances,9 acid substances,7 ester substances,4 alcohol substances and 3 other types of substances.Among them,the content of 1%or more is?Z,Z?-9,12-octadecadienoic acid,2-ethylhexyl phthalate,9,17-octadeca diene,trans Squalene,?-sitosterol,dibutyl phthalate,n-hexadecanoic acid,stearic acid,arachidic acid,campesterol,accounting for 90.74%of the total peak area.Squalene and acid campesterol that are detected all have certain anti-tumor activity.2.The chemical composition and anti-tumor activity of different polar solvent extracts were studied.The extracts of different polarity solvents were analyzed by High Performance Liquid Chromatography?HPLC?.At the same time,the anti-tumor effect of n-hexane extract and chloroform extract on H22 subcutaneous tumor-bearing mice was investigated.Ethyl acetate extract,n-butanol extract and ethanol extract have nhibitory action on human hepatocellular carcinoma cell line HepG2by MTT method.The results showed that the best anti-tumor effect is from ethyl acetate extract,400?g/mL of ethyl acetate extract inhibited 48h up to 63.07%,compared to the chromatogram obtained by HPLC.It could preliminary inferred the possible active ingredient is 16 min and 34min out of the peak material,which provides a theoretical basis for the follow-up screening of active ingredient,separation and enrichment.
