Regarding periodontal disease, the incidence of gingivitis and gingival bleeding is high.As daily cleaning and caring is an important and effective means to prevent oral diseases, correct tooth-brushing will help ensure oral cavity health considerably. The effect of the toothpaste has evolved from single cleaning to a variety of auxiliary therapeutic properties. Therefore, the toothpaste is formulated with additional active ingredients so as to achieve the antibacterial and anti-inflammatory effects. This will help reduce plaque, bacteriostasis, inflammation and it will also assist the treatment of gum bleeding and periodontitis arising from partial infection.Hence it proves very necessary to find out safe and anti-inflammatory active ingredients to be applied in the toothpaste. There are three commonly used methods: clinical trial evaluation, laboratory test evaluation and animal test evaluation.Prior to clinical trials, two methods of laboratory test evaluation and animal test evaluation are often adopted inorder to improve the efficiency of product research work and to save the cost of clinical trials.During the laboratory test evaluation, the most common way is to test the toothpaste for bacteriostatic rate test against specific pathogens. For this study, the dominant pathogenic bacteria from the subgingival gum bacterial plague--gum porphyromanus gingivalis as typical pathogenic bacteria causing gingivitis is adopted for bacteriostat experiments.Invitro simulation of animal experiments, the mouse ear swelling inflammatory casewas selected to predict the anti-inflammatory activity of active ingredients. This study covers the experiment of dipotassium glycyrrhizic used in toothpaste. The experiment result has showed the toothpaste containing dipotassium glycyrrhizic could produce the antibacterial activity against the oral inflammatory bacteria. The study has experimented on the MIC(minimum inhibitory concentration)of dipotassium glycyrrhizic against gum porphyromanus gingivalis. This study has covered the research on how a toothpaste containing 0.2% dipotassium glycyrrhizic wil be inhibitive against gum porphyromanus gingivalis and how a p-xylene will affect the mouse auricle swelling. The experiment results shows that dipotassium glycyrrhizic is inhibitive against gum porphyromanus gingivalis with MIC 1.875g/L,a toothpaste containing 0.2% dipotassium glycyrrhizic wil be inhibitive against gum porphyromanus gingivalis and the bacterial inhibition ring diameter 20.5mm;Based on the anti-inflammatory pattern of p-xylene causing mouse auricle swelling, the toothpaste with 0.2% dipotassium glycyrrhizic features super positive comparing to dexamethasone acetate paste..Finally,high performance liquid chromatography(HPLC)was used to verify the concentration of the toothpaste containing dipotassium glycyrrhizicConclusion: this study has verified that that the toothpaste containing dipotassium glycyrrhizic is workable for typical pathogen arising from oral inflammation-,porphyromonas gingivalis bacteriostasis and for decreasing of mouse auricle swelling rate induced by p-xylene,While through invitro experiment. It showed that the toothpaste of this type will be preventive against gingivitis.
