| Objective:To synthesize the polyethylene glyco(PEG)-grafted chitosan coupled with deoxycholic acid,and to prepare the Curcumin-loaded micelles and further characterize their physicochemical properties.Method:(1)PEG-chitosan-deoxycholic acid was synthesized by two steps,and its structure was characterized by IR,1H NMR and elemental analysis on the intermediate products and final products.The physical properties were charaterized by wide angle X ray diffraction(WAXD)and differential scanning calorimetry(DSC).The solubility of carrier was investigated in various solvents,and the solubilization capacity for insoluble drugs was also discussed.(2)The preparation method of micelles was optimized,and the entrapment efficiency,drug loading,particle size distribution and drug release behavior were also characterized.Result:(1)The grafting ratio of PEG was 11%,the intrinsic viscosity of deacetylation chitosan was 67.38 mL.g-1,the intrinsic viscosity of mPEG-CS was 27.913 mL·g-1;(2)the critical micelle concentration of the carrier material was 5.236*10-5 mol·L-1;the drug loading amount of the Curcumin-loaded micelles was(12.11 ± 1.52)%,the encapsulation efficiency was(89.37±4.12)%,and the average particle size was 161.4nm;the drug release from the micelles was relativley slow.Conclusion:A new type of chitosan derivative with amphiphilic charateristics was synthesized in this study.It will be an excellent micelle carriers.It showed high encapsulation efficiency and drug loading for curcumin. |
