25 groups of different concentrations ratio of PAEs(phthalic acid esters)and BPA(Bisphenol A)were blended to process Glycine max seeds,and determine the changes of malondialdehyde content,soluble sugar content,superoxide dismutase activity,peroxidase activity,soluble protein content and free proline content.Through the total RNA extraction from Glycine max seeds,reverse transcription synthetic cDNA first chain,related primer amplification and clone of HSP70 part sequences homologous sequence alignment,HSP70 gene containing in Glycine max seeds were determined.Western Blot was used to detect protein expression levels of HSP70 genes in the process of Glycine max seeds development.RT-PCR techniques were utilized to detection the expression differences of HSP70 gene under the stress of different concentration of environmental hormone PAEs and BPA combined pollution.The experimental results showed that the changes of malondialdehyde content and soluble protein content in Glycine max seeds have the trends of decreasing first and increasing then under the condition of PAEs,BPA single pollution.Soluble sugar content,superoxide dismutase(SOD)activity,and peroxidase(POD)activity increased first and decreased late.Free proline content increased more and more.And under the condition of PAEs,BPA combined pollution,malondialdehyde content and peroxidase activity in Glycine max seeds increased after the first reduce trends.Soluble sugar content,SOD activity,free proline content,and soluble protein have the trends of decreasing first,and then increase.This experimental result of gene clone sequences showed that the amplified fragment belongs to heat shock protein 70 gene fragment.Sequences alignment results of 24 experimental groups showed that PAEs and BPA pollutions have mutagenesis on HSP70 gene and this kind of hazard function of combined pollution is larger than that of single pollution.RT-PCR and western blot results showed that quantitative detection of RT-PCR and qualitative detection of western blot test results are consistent.That was the expression quantity of HSP70 gene and protein increasing first and decreasing late.Compared with the single environmental hormone pollution,the effects of combined pollution of environmental hormones on HSP70 in Glycine max seed show that low concentration promote and high concentration inhibit.
