Heat shock proteins(HSPs) is ubiquitously distributed and the importantly isoforms heat shock protein 70 (HSP70) is strongly induced by environment, such as heat, heavy metals, inflammatory cytokines, UV irradiation organic materials and heavy metal.Thus, HSP70 is an effective biological marker for reflecting the exist and extent of environmental pollution. It plays an important role in environmental monitoring and environmental protection. In this study, carp was used as experiment material and HSP70 was extracted from carp by purification and isolation methods, such as tissue breaking, salting, chromatography and electrophoresis. The specific sequence of affinity ligand of HSP70 was obtained with the 12 Phage Display Peptide Library subsequently. After three rounds of panning, the affinity ligand sequence of HSP70 was obtained. The biological probe was synthesized by manual synthetic peptide way. Under the condition that the carp was subjected to Cu2+ stress, we detected the carp serum HSP70 expression level by the biological probe. A new method for rapid detection of water quality was established which will give some services for environmental monitoring and environmental management. |
