Kenaf stalk degumming is preparing based and key technology of fiber pretreating process,degumming of good or bad will directly affect the fiber quality and quality.However,screening to the strains of degumming,poor ability of single type of enzyme production,so as to make the kenaf fiber degumming is not complete.Therefore,our main task is variety breeding to produce enzymes,high enzyme activity of kenaf degumming strains.Based on pieces of kenaf bast as inducer,After early screening can get thirty in kenaf bast as the sole carbon source of culture medium on growth of strain.The single colony after screening,selected can produce larger transparent circle on pectin medium plate of single colony,and access to the degumming medium can make kenaf bast degumming dispersed in fibrous,is also one of high yield and pectin enzyme strain.By classic methods for 11-2 bacteria identification,determination of strain of physiological and biochemical reaction,determination of strain for the 16S rDNA sequences,and identification of fatty acid,and finally determine the strain bacillus Paenibacillus for class.11-2 strains enzyme degumming of extracellular enzyme production,enzymology properties study,pectic enzyme reaction temperature of 40~60 ℃5 the optimal reaction temperature is 50 ℃;Enzyme reaction pH between 8.0 and 10.0,the optimum pH is 9.0.Metal ion impact experiments show that the Ca2+ and Mg2+ for the activation of enzyme reaction has the obvious effect,Zn2+ and Fe2+ effect is not obvious,Cu2+ and Mn2+ has obvious inhibitory effect.11-2 in kenaf based on strain fermentation medium middle pectic enzyme activity of 20167.95 U/mL,xylanase activity of 364.85 U/mL,through the initial pH of the fermentation medium,fermentation temperature,fermentation time and quantity of optimized fermentation condition optimization,11-2 strains in the pH value is 9.0,the temperature is 37 ℃,time of 24 h,inoculation quantity was 5%under the optimum fermentation conditions of,pectic enzyme activity reached 40643.30 U/mL,before optimization is 2 times less than it,the pH value is 5.0,the temperature is 37 ℃,time of 24 h,5%inoculated quantity the best fermentation conditions,the xylan enzyme activity reached 554.27 U/mL,more than doubled before optimization.
