Konjac is a kind of perennial herbaceous plants,bulbs are rich in large amounts of dietary fiber Konjac glucan-mannan chitosan(Konjac glaucomannan,KGM).KGM has the effect of hpyerglycemic,hypolipidemic,anti-cancer,adjusting the gastrointestinal tract and regulating immunity.KGM has the very strong water absorption swelling and contains no calories,It is also difficult to digestion and absorption,so as to prolong the residence time in the stomach cavity and intestines,slow the digestion of food,which makes people feel fuller.At the same time It can improve insulin sensitivity,reduce glucose tolerance,To be the first choice of the person with diabetes health care products.KGM with other polymer gel can produce synergy to improve the properties of a single gel,which can be used in the capsule wall material to make it has high degree of swelling in the intestines and slow-release effect in stomach.Apple,is the main fruits in China,and its taste sweet,rich nutrition.apple polyphenols have the function of oxidation resistance,anti-cancer,dental caries prevention,hypotensive and hypolipidemic.At present,the most widely used is tea polyphenol,apple polyphenol application study is less,but it has a higher capacity to remove free radicals than tea polyphenolThis research is mainly through the different concentrations of ethanol washing konjac powder to make purified powder.apple polyphenol was extracted with ultrasonic auxiliary method from Apple residue By single factor test and four factors three levels orthogonal experiment,finally determine the optimal extraction process.Using the AB-8 macroporous resin to adsorb apple polyphenol.then draw the dynamic adsorption curve,determine the volume of collection of eluent,get the purification of apple polyphenols solution.Analysis the purification apple polyphenols by HPLC.compared with 11 kinds of mixed standard,through qualitative test,determine the free phenol.Spectrophotometric method is used to analyse the influence of free phenol and different concentration of Polyphenols liquid on the ACE inhibition rate.Using the swelling property of purified KGM,the sustained-release capsules can enter the gastrointestinal tract,slow swelling and at the same time prevent polyphenols rapid dissolution,conducive to the stability of polyphenols.Relative to the general system of chitosan,sodium alginate sustained-release capsules,It has better slow-release effect.By single factor test and orthogonal test,the capsule formulation was optimized.Studied on the influention of proportion of KGM and sodium alginate,Cacl2 concentration,chitosan concentration,polyphenols and total sol on the swelling degree in simulated intestinal fluid and the release rate of polyphenols in simulated gastric juice,by four factors three levels orthogonal experiment,finally determined the optimal formula..Finally to evaluate slow-release effect,swelling degree and light,oxygen,temperature stability of the capsule.The results showed:1?After ethanol purification,to get the average level in 69.80%of KGM in konjac purified powder.2?By single factor screening of extracting conditions and the four factors three levels orthogonal experiment,finally get the best extraction technology for:ethanol concentration of 60%,and solid-liquid ratio of 1:10,ultrasonic power:350 w,ultrasonic time:40 min.Under this condition,apple polyphenols extraction yield is 3.721 mg/g.Use 60%ethanol to elute AB-8 macroporous resin which adsorption saturation,then collect the liquid product in 30?60 ml,which apple polyphenol content is the highest.3.Through the HPLC analysis,.the polyphenols contains epicatechin,quercetin,quercetin and caffeic acid,From the hypotensive test in vitro,they all have the ability to control ACE enzyme.The inhibition effect is epicatechin>quercetin>quercetin>caffeic acid.Do the test of hypotensive in vitro by Polyphenols solution,the results show that with the increase of polyphenols extract concentration,the inhibition rate increased gradually,the polyphenols concentration in 200 ?g/ml to 350?g/ml,Inhibition rate remain unchanged.4.Through single factor experiment and four factors three levels orthogonal experiment,finally got the best formula is:the proportion of KGM and sodium alginate:1:4.5,the ratio of polyphenols and total sol:1:9,the concentration of CaCl2:1.5%,chitosan concentration:0.5%5.Through the capsule slow-release effect evaluation,glucan-mannan chitosan/chitosan/sodium alginate capsules made its slow-release effect better than that not to join the KGM,that is to say chitosan chitosan/alginate sodium system.And its stability under the condition of air,temperature,light is very good,and it is better than that of polyphenols power.
