| In order to making full use of apple pomace and getting the much higher purification of apple polyphenols,we taked the agricultural waste material—apple pomace as the research material in this research.,with gallic acid as standard sample, the optimum conditions for determining of the polyphenols in the apple pomace by Folin Method were investigated; Microwave assisted extraction technique of Apple polyphenols in apple pomace was studied by quadratic perpendicularity and universal rotational designs, the optimum conditions was determined; Using polyphenols of apple pomace from microwave assisted extraction to contrast with usual antioxidant to determine the effects on antioxidation to fats and Peanut oil; Discussed the optimum conditions for macroreticular purification of AP; Researched the technological parameters of purification conditions of AP by Sephadex LH-20; Some active contents in polyphenols was identified by using HPLC method. The conclusions of this research were as follows:1,The results showed that the optimum reaction conditions were volume fraction of Folin reagent and 10 %Na2CO3 1:1 (v:v), reaction temperature 25 °C, reaction time 120 min., .The average recoveries were 106.5 %, the RSD was 0.49 %. It had a good accuracy and precision to determine the polyphenols content.2,The optimum conditions were extraction time 60 S, microwave radiation 750 W , ethanol concentration 60 %,the rate of solid to solution 1:50 , the maximum of absorbency was 0.460; the extraction rate of Apple Polyphenols was 11.93 mg/g(DW) in theory, it is higher than that of refluxing and ultrasonic extraction method.3,AP acted well in anti-oxidation on peanut oil and fats.4,Different resins performed differently in adsorbing AP; and NKA-9 was the most appropriate in separating and purifying of AP among the four chosen macroreticular resins. When the concentration of AP was 2.5 mg/mL, 60% ethanol used as eluent at the speed of 1.0 mL/min, the purity of AP could reach the height of 19.74%, which was approximately 20 times higher than the previous0.97%, the yield of AP could reach 66.34 %. |
