Objective:In recent years,stem cells have been widely concerned in the field of tissue engineering and regenerative medicine due to their advantages of abundant source,simple sampling and low immunogenicity.A large number of studies have shown that exosomes derived from stem cells have the functions of promoting tissue repair and regeneration,and can prevent immune rejection caused by stem cell transplantation.Therefore,stem cells derived exosomes have a great prospect in the field of tissue repair.Thus,this paper studies the effect of adipose stem cell derived exosomes on keratinocytes,aiming to provide theoretical basis for regulation of tissue regeneration and repair by exosomes derived from stem cells.Methods1.Firstly,the subjects related to the test were informed to the healthy parturient of the second cesarean section aged 18-30 years with previous scars,and the informed consent was signed.The discarded subcutaneous tissue was collected,and the adipose stem cells were extracted and identified under the electron microscope.2.Inform the adult male aged from 18 to 25 about the relevant test matters,sign the informed consent,collect the discarded foreskin tissue,isolate the keratinocytes and identify them under the electron microscope;3.Exosomes derived from adipose stem cells were extracted by SBI reagent,and exosomes were identified by Nano FCM Flow Nano Analyzer and western blot.4.Different concentrations of exosomes derived from adipose stem cells applying to keratinocytes,and using the scratches,CCK-8,transwell method to detect cutin cell proliferation and migration by RT-PCR and ELISA kit method on cutin cell secretion of MMP-1(matrix metalloproteinases-1),Il-1?(interleukin-1?),TGF-?(transforming growth factor-?);5.The mouse trauma model was constructed,and the wound site was embedded with exosomes derived from adipose stem cells,and finding the changes on day 3,5,7 and 14 of MMP-1(matrix metalloproteinase-1)and MMP-3(matrix metalloproteinase-3)by immunohistochemical staining.Result1.Exosomes derived from adipse stem cells can promote the proliferation and migration of keratinocytes;2.Exosomes derived from adipse stem cells can promote the secretion f MMP1-1,Il-1? and TGF-?3.In animal experiments,exosomes derived from adipose stem cells acting on mouse trauma sites,the experession of MMP-1 and MMP-3 were incresed.Conclusion: Exosomes derived from adipse stem cells can promote the proliferation and migration of keratinocytes,regulate the expression of secreted MMP1-1,Il-1? and TGF-?,and promote the expression of MMP-1 and MMP-3 in vivo.
