ABA(Abscisic acid)is a key stress hormone which regulates morphological and physiological aspects of plant growth and development.A number of genes have been reported to be involved in ABA signaling and stress regulation in Arabidopsis thaliana,however,functions of some ABA responsive genes still remained unidentified.In this study,we report the identification of AWR(ABA repressed WD40)as ABA responsive gene in Arabidopsis.Quantitative RT-PCR results revealed that the expression levels of AWR decreased significantly in response to ABA treatment.Protoplast transfection assays showed that AWR is located in cytoplasm and cell membranes.RT-PCR analysis showed that AWR highly expressed in stem but it is also expressed in other organs such as Root,rosette leaves and stem leaves.Seed germination analysis revealed that AWR overexpression plants were more sensitive to ABA.Overexpression plants were reported to be germinated faster than Col wild type and mutant respectively at any given concentration.Cotyledon greening assay showed that 35S:AWR transgenic plants were more sensitive to ABA as compared to Col wild type and mutant plants.Na Cl assay showed that AWR overexpression plants were more sensitive to salt stress as compared to Col wild type and mutant plants.Observation of morphological phenotypic revealed that AWR overexpression transgenic plants have longer roots as compare Col wild type and mutant plants in control.Root elongation essay also showed that overexpression plants have longer roots as compared to other lines in ABA.It was reported that overexpression plants were more sensitive to NaCl treatment in root elongation essay.In response to Mannitol overexpression lines showed longer roots than the Col wild type and mutant lines.In early stages the growth of AWR overexpression plants were faster than Col wild type and mutant plants.Altogether,our results suggest that AWR is involved in the regulation of ABA response and plant growth and development in Arabidopsis.
