With the development of living standards,the probability of cardiovascular diseases has increased year by year.One of the leading factors to cardiovascular diseases is high cholesterol level.Several academic articles have shown that probiotics can improve immunity,balance intestinal flora,regulate blood lipids,decrease blood pressure and blood sugar,as well as anti-cancer and anti-tumor.Probiotics including Lactobacillus rhamnosus(BMBL0606),L.reuteri(BM BL0612),L.casei(KJ598784),L.acidophilus(BMBL0618),Bifidobacterium.Lactis(BMBL0624),B.animal(BMBL0630)were selected to study.Two probiotics with obvious cholesterol-lowering effect in vitro were selected through the evaluation of the reduction effect of cholesterol in vitro.The evaluation consisted of three parts,the reduction of cholesterol in vitro,the precipitation of eluent cholesterol and the activity of bile salt hydrolase.High-fat mice were fed to explore their cholesterol-lowering effects in vivo.The experimental results were as follows1)Total cholesterol clearance in vitro: When being cultured for 12 hours,the cholesterol clearance rates of the probiotics in vitro was 49.80%,51.50%,46.47%,48.67%,37.47%,and 51.35%.The clearance efficacy was 8.17,18.61,15.50,17.58,10.41,and 11.13 ?g/ m L g.2)Effect of co-precipitation: There were no significant differences in the cholesterol content of the precipitated eluate between the strains,ranging from 1.91-2.76 ?g / m L.Six probiotics reduced cholesterol in vitro mainly by assimilation.3)Activity of bile salt hydrolase: All probiotics had bile salt hydrolase.BMBL0612 and BMBL0618 showed higher enzyme activity.The total enzyme activites of BMBL0612 in sodium glycinate,sodium taurocholate,and mixed bile salts were5.845,5.309 and 4.465 units(?mo L m L-1 min),and the specific enzyme activities were 0.080,0.073 and 0.061 units(?mo L ?g-1 min-1).The total enzyme activities of BMBL0618 in sodium glycinate,sodium taurocholate,and mixed bile salts were5.921,5.309 and 3.393 units(?mo L m L-1 min),and the specific enzyme activities were 0.146,0.130 and 0.084 units(?mo L ?g-1 min-1).4)Evaluation of cholesterol-lowering effect in vivo: The intervention of probiotics could inhibit the weight of mice and reduce the body fat ratio to a certain extent.The abnormalities of triglyceride(TG),total cholesterol(TC),and low-density lipoprotein cholesterol(LDL-C)levels in the blood of high-fat mice in each intervention group were all improved.The triglyceride content returned to the normal range.The overall effect of BMBL0618 was better than that of BMBL0612.5)To sum up,this article took six kinds of probiotics as research objects.All six kinds of probiotics showed cholesterol clearance ability.BMBL0612 and BMBL0618 with high cholesterol lowering ability in vitro were screened through comprehensive evaluation of cholesterol lowering in vitro.Compared with the in vivo cholesterol-lowering experiments represented by BMBL0612 and BMBL0618,BMBL0618 had better comprehensive ability on blood lipids between high-fat mice.The results provided a theoretical basis for the development of probiotics cholesterol-lowering functional foods.
