The Viral Metagenomic method was used to construct a Metagenomic library of fecal virus in forest musk deer and elk deer,analyze the virus communities in their fecal samples,and dig out new viruses and analyze their genetic characteristics.The main contents and conclusions are as follows:1.Analysis of enterovirus communityThe enterovirus community of forest musk deer mainly includes Circoviridae(12.6726%),Microviridae(42.7562%),Mimiviridae(1.2180%),Nodaviridae(5.7245%),None(25.8715%),Phycodnaviridae(2.5980%),Siphoviridae(2.1163%),Others(7.0429%).There are many viruses capable of infecting vertebrates,among which Astroviridae,Circoviridae,Nodaviridae and Picornaviridae constitute the main part,and Circoviridae accounts for the largest proportion.The enterovirus community of forest musk deer mainly includes Circoviridae(12.5612%),Microviridae(36.1924%),Mimiviridae(3.6175%),None(33.7381%),Phycodnaviridae(3.7572%),Siphoviridae(5.5346%),Others(4.5989%).There are a variety of viruses with the ability to infect vertebrates,such as Circoviridae,Herpesviridae,Marseilleviridae and Poxviridae,etc.Comparing the enterovirus communities of forest musk deer and elk deer,it was found that there was little difference in the number of virus species between the two groups,and There are only minimal differences in the types of viruses.2.Identification and genetic characteristics analysis of novel CRESS-DNA virusNine strains of CRESS-DNA virus were found in the intestinal tract of forest musk deer.Blastp homology analysis showed that UJSL001(3518 nt,Lin2),UJSL003(3212 nt,Lin3),UJSL006(2148 nt,Lin7)and UJSL017(3025 nt,Lin1)were homologous to CRESS-DNA virus,with a similarity of 44.00% to 62.54%.UJSL002(2665 nt,Lin3),UJSL004(2866 nt,Lin5)and UJSL007(2526 nt,Lin9)maintained high homology to smacovirus,with similarity of 69.09%-95.54%.ls-cyc(3552 nt,Lin7)and UJSL005(3852 nt,Lin6)maintained high homology with Circoviridae,with similarity of 34.31% and 32.63%,respectively.Six strains of CRESS-DNA virus were found in elk intestinal samples with homologous similarity ranges of 41.05%-84.49%,which were Elk06cre1(2036 nt,Elk06),Elk06cre2(3373 nt,Elk06),Elk08cir(2431 nt,Elk08),Elk10cre1(2275 nt,Elk10),Elk10cre2(2772 nt,Elk10)and Elk10cre3(2657 nt,Elk10),respectively.3.Identification and genetic characteristics analysis of other vertebrate virusesThe sequences of nine vertebrate viruses were analyzed in the intestinal tract of forest musk deer,belonging to the family Astroviridae,Parvoviridae and Picornaviridae.Twenty virus sequences were analyzed in the intestinal tract of elk,belonging to Arteriviridae,Bunyaviridae,Caliciviridae,Coronaviridae,Flaviviridae,Filoviridae,Orthomyxoviridae,Paramyxoviridae and Picornaviridae.Conclusion: All the virus communities in the intestinal tract of forest musk deer and elk were revealed by means of virus metagenomics,and the differences between the two virus communities were not obvious.Nine and six new strains of CRESS-DNA virus were found in the intestines of forest musk deer and elk deer respectively.Homology and phylogenetic analysis of other vertebrate viruses in the intestinal tract of forest musk deer and elk were carried out to identify the category of these viruses.
