Objective To explore the effects of interleukin-2(IL-2),interleukin-15(IL-15)and their combinations on the number and function of primary mice natural killer(NK)cells.Method The purified NK cells were obtained from mice spleen mononuclear cells by immunomagnetic separation.The number of living cells was calculated by trypan blue staining.The cells were divided into IL-2 group,IL-15 group and IL-2 combined with IL-15 group.The effects of different concentrations of cytokines on the expansion of mice NK cells in vitro were studied.Using YAC-1 / GFP cells as the target cells,the killing activity of NK cells was detected by flow cytometry.Result 1.Adding different concentrations of IL-2 for 24,48,and 72 hours of culture.The number of NK cells expanded in the low,medium,and high concentration groups is greater than that in the control group at different time points,and there are statistical differences between the groups(P < 0.05),the expansion of NK cells does not increase with time.At the same time point,the expansion capacity of NK cells increased with increasing concentration(P <0.05).2.Adding different concentrations of IL-15 for 24,48,and 72 hours,the low-concentration group for 24 and 48 hours was statistically significant compared with the control group(P<0.05).The medium-and high-concentration groups had different levels of NK cells at different time points.The number of expansions was greater than that of the control group(P<0.05).The number of NK cells expanded with the increase of concentration at 24 and 48 hours of culture,and there was a statistically significant difference between the different concentration groups(P <0.05).3.After determining the optimal concentration,the number of NK cells expanded in the IL-2+ IL-15 combination group was greater than that in the control group at different time points(P <0.05).There was no statistically significant difference between the experimental groups at24 and 48 hours of culture,and there was statistical significance at 72 hours of culture between IL-2 group> IL-2 + IL-15 group> IL-15 group> Control Group(P <0.05).4.In the killing experiment of NK cells,the effect of IL-15 and IL-2 + IL-15 on YAC-1 / GFP cells was higher than that of the control group(P < 0.05).When the effective target ratio was5:1,NK cells had the highest killing activity,which was IL-2 + IL-15 group > IL-15 group >control group(P < 0.05).When the effective target ratio was 10:1,there was no significant difference between IL-2 + IL-15 group and IL-15 group(P > 0.05).Conclusion 1.Different concentrations of IL-2,the higher the concentration,the greater the number of expansion of mice NK cells;2.Different concentrations of IL-15,the higher the concentration,the more the expansion of mice NK cells;3.IL-2 Combined with IL-15 can enhance the in vitro expansion capacity of mice NK cells;4.IL-15 and IL-2 combined with IL-15 can enhance the killing activity of mice NK cells on YAC-1 / GFP cells,and the effect-target ratio When it is 5: 1,the NK cell killing activity of the IL-2 + IL-15 group is the strongest,followed by the IL-15 group.
