Parkinson's disease is a common neurodegenerative disease known as "silent killer".The destructing process from this disease is slow and irreversible.At present,the condition can only be improved without cure.While the mechanism of FOG remains uncertain,most studies were based on brain network instead of separate brain areas.Only a few studies investigated the functional magnetic resonance imaging(f MRI)with gait-freezing symptoms.Studying the relationship between large-scale whole-brain functional connectivity and phenotypic factors may help reveal the nature of FOG symptoms.The samples in this study are divided into three groups: 47 patients with self-reported gait freezing symptoms(PD+FOG),28 patients without FOG(PD-FOG),and 28 healthy people as a control group(HC).Clinical scale assessment,behavioral assessment and functional magnetic resonance imaging were performed on these three groups.The emergence of a multivariate statistical method called Multiple Distance Multiple Matrix Regression(MDMR)enables regression analysis without a prior hypothesis or false positives which are often observed in multiple comparison.Based on the original f MRI image,this study utilizes a functional mapping technique,which includes 264 brain nodes in the whole brain,as the critical method to perform segmentation.Using MDMR-CWAS to find statistically significant brain nodes,ie,seed points.We rank each seed point based on effect level and compares them with other nodes on total intensity of the whole brain connection.Combining the graph theory parameter analysis such as local node efficiency and global small world,researches are done on the inter brain network functional strength differences between groups.The results shows that Group PD+FOG has worse clinical manifestations and behavioral assessment.Based on diagnosis,Mo CA,delayed recall and step count tests,five statistically significant nodes allocating in sensation motion network and visual network are discovered.Based on a discrete scale such as a diagnosis,the significant brain nodes are located in the central back of the perceptual motion network.This paper finds that differences between normal people and Parkinson brain connection patterns are common.Differences between the Parkinson subgroups are mainly due to changes in the composite pattern between the cerebellar network and the subcortical network and related networks.
