Both Cephalotrichum and Scopulariopsis are classified into Sordariomycetes,Microascales,Microascaceae.Species of Cephalotrichum and Scopulariops are saprophytic or weakly parasitic fungi which are widespread in soil and also can be isolated from air,plant debris,animal waste,textiles,cheese,and wet indoor environments.some species of two genera serve as indoor fungus which can infect human being and threaten human health.They are also important plant pathogens which can infect many kinds of plants.On the other hand,some species of the two genus fungi can produce a variety of beneficial secondary metabolites,which can be used in medical,industrial and agricultural fields.Therefore,the accurate identification of the two genera is the basic for further research and development.By 2017,Index fungorum recorded 81 species of Cephalotrichum and 112 species of Scopulariopsis.Although there are many kinds of records in the two genera,due to lack of some model specimens and traditional morphological identification methods insufficient,taxonomic status of the two genera that are controversial and constantly changing.From 2017 to 2019,more than 600 soils samples were collected from Guiyang,Luodian,Zunyi and other places in Guizhou Province,and more than 400 strains were isolated by plate dilution method.11 strains of Cephalotrichum were identified.In addition,Shandong Agricultural University donated 44 strains of Cephalotrichum and 13 strains of Scopulariopsis.Therefore,a total of 55 strains of Cephalotrichum and 13 strains of Scopulariopsis were obtained.After morphological studies and phylogenetic analyses with combined ITS,LSU,TUB2 and TEF1 gene regions,4 new species,1 new record species in China and 7 reported spices are identified for Cephalotrichum.New species are C.catenulatum,C.humicola,C.tongrenense and C.septum;new record species in China is C.domesticum;reported species are C.asperulum,C.gorgonifer,C.microsporum,C.nanum,C.stemonitis,C.verrucisporum,C.purpureofuscum.In Scopulariopsis and its sexual gengus Microascus,2 new species,namely M.ellipsoideum and M.globose;1 new combination species,M.verrucifera;1 new Chinese record species,M.verrucosus;2 reported species,S.asperula and M.croci were identified.To select and evaluate gene fragment of Cephalotrichum and Scopulariopsis,we built mono and muli-locus MP?ML?BI phylogenetic trees with ITS,LSU,TUB2 and TEF1 gene region respectively.Using three algorithms of maximum likelihood(ML),maximum parsimony(MP)and Bayesian inference(BI)built mono and muli-locus phylogenetic trees were analyzed to distinguish Cephalotrichum,Scopulariopsi and their approximate genera,And combined with the amplification rate and support rate,select and evaluate optimal gene fragments suitable for two genera.
