Fundamental Study On Biological Mechanism Of Photomagnetical Reception In Pigeons’ Retina

Abstract/Summary:
The pigeon’s navigation ability is very outstanding,and its biological mechanism of relying on geomagnetic navigation has attracted widely attention in recent years,but the mechanism remains unknown.In this thesis,the magnetic-reception behavior of pigeons under different conditions was investigated by optimizing combined techniques,such as performing experiments on molecular biology,cell electrochemistry and cell electrophysiolog.Some novel magnetic navigation mechanism of pigeons were revealed.Hence,how the pigeon retina participates in the photomagnetic response under different conditions is obtained.Firstly,the cryptochrome mRNA level under varied conditions was analyzed by transcriptosome sequencing.Afterwards,quantitative real time-polymerase chain reaction(qRT-PCR)was performed to further validate the mRNA level of cryptochrome.Owing to transcriptosome sequencing stastics,the transcriptosome in pigeon was authenticly diversed under only the blue light irradiation.Besides,the commonly dissimilarities were contributed in phototransduction pathway,periodic rhythm pathway.Then,qRT-PCR was performed and resulted in that the cryptochrome 1 had significant fold-change under blue light irradiation,which is unanimous to the existing reports.Then,a bionic-dove’s retina photomagnetic sensing system based on nanocomplex of gold nanoparticles and cellulose was resembled to detect whether the retina could respond to photomagetic stimulation or not.The impedance of the whole sensing system changes while the external magnetic field exists,indicating that the retina can respond to the magnetic field.Moreover,the blue light stimulation could similarly solicit the impedance hiking,unlike red light stimulation.Together,these results demostrated dove’s retina could respon to photon stimulation and eventually results in impendence change.Furthermore,sinle sensillum recording system was used to varify the photomagnetic response of doves’ s retina.The response to blue and red light of dove’s retina could be detected with using SSR single sensillum recording system,while the response was not significant,but a significant response of the dove’s retina to the magnetic field is detected.These results may pave the way towards umderstanding the mechanism of photomagnetic reception of pigeon.
