The subfamily Agrilinae belongs to the family Buprestidaethe of the superfamily Buprestoidea in the order Coleoptera.There are 4 tribes 23 subtribes and 200 genera in the subfamily Agrilinae.The previous research of Agrilinae mainly focused on the traditional taxonomy,and there were few reports on the research of molecular phylogeny.At present,the classification system of the subfamily Agrilinae is more controversial,and the phylogenetic relationship of the subfamily Agrilinae is not clear.This paper provide here a molecular phylogeny of the Agrilinae,chosing 4 Bupretinae species within 4 genera and 1 Polycestinae species within 1 genus as outgroup,69 species of Agrilinae from 20 genera of 4 tribes in 13 subtribes,as ingroup.Based on a combination of mitochondrial and nuclear markers(COI,Cytb,18 Sr DNA and28 Sr DNA),the phylogenetic trees were constructed using Maximum Likehood(ML)and Bayesian method.The systems at the rank of tribes were discussed in the subfamily Agrilinae.In this study,51 COI sequences,57 Cytb sequences,41 18 Sr DNA sequences and 31 28 Sr DNA sequences were obtained by self sequencing,totaling of 180 sequences.The phylogenetic tree constructed by the four-gene joint dataset is found more stable in topological structure and higher confidence coefficient than the three-gene joint dataset,and the Bayes tree is more similar to the Maximum likelihood tree.In the established phylogenetic trees,the basal topology of Bayes tree and Maximum likelihood tree are different,the topological structures of most terminal branches ares the same,the confidence value of nodes are different and Bayes tree is more paraphyletic clades.The high confidence value of the branch proves the reliability of the branch topological structure.The following conclusions are made based on the phylogenetic tree constructed by four gene sequences:1.The subfamily Agrilinae is a monophyletic group.2.The tribes Coraebini,Aphanisticini,Tracheini and Agrilini are polyphyletic groups.3.The subtribe Leiopleurina is a monophyletic group.The subtribe Trachyina is a polyphyletic group.4.The genera Sambus,Callimicra,Agrilus,Nalanda,Pachyschelus,Coraebus,Aphanisticus,Taphrocerus and Habroloma are monophyletic groups.The genus Paracylindromorphus is a polyphyletic group.The genera Trachys and Leiopleura are paraphyletic groups.5.The genus Leiopleura is a sister of the genus Callimicra,and they are closely related.6.The genus Nalanda(The subtribe Meliboeina)is a sister group to the genus Pachyschelus(The subtribe Pachyschelina),and they are closely related.7.The genus Coraebus(The subtribe Coraebina)is a sister group to the genus Aaaba,and they are closely related.8.The genus Trypantius(The subtribe Dismorphina)is a sister group to the genus Meliboeithon,and they are closely related.9.The genus Trachys is a sister group to the genus Habroloma,and they are closely related.10.The genus Neotrachys is distant from other genera of the subtribe Trachina,this paper suggests that the genus Neotrachys should be removed the subtribe Trachina and should be included in the tribe Aphnaisticini.11.The genus Trypantius(The subtribe Dismorphina)and the genus Meliboeithon form a cluster,and the taxonomic status of the genus Meliboeithon is undetermined.This paper suggests that the genus Meliboeithon should be included in the subtribe Dismorphina.
