Paenibacillus glucanolyticus is a strain isolated from pure water for production,and the detection rate of the strain in pure water is high for a period.If the final production is polluted by pure water,bacillus is easy to form nutritional cells in the food,which threatens the safety of the product.In order to ensure the quality of pure water and products,the target bacteria must be effectively killed.First studied its colony morphology and physiological and biochemical characteristics,and then through the analysis of several killing methods,compared of its efficacy,advantages and disadvantages in killing Paenibacillus glucanolyticus to find out the effective killing methods and conditions.Through practice,found that the compound disinfectant of peracetic acid has a good killing effect on Paenibacillus glucanolyticus.First,the morphology of the colony isolated from pure water for production was observed,and it was found that the cell was rod-shaped,there were elliptic spores in the expanded cell capsule,and the Gram staining was negative.According to its physiological and biochemical characteristics,it is preliminarily determined that the strain is Paenibacillus glucanolyticus.Physiological and biochemical identification includes anaerobic,motility,growth curve drawing,contact enzyme test,oxidase test,gelatin liquefaction,deoxyribonuclease test,casein hydrolysis,cellulose hydrolysis,and other tests.Then the 16 S r DNA sequence was used to identify the strain.The results showed that the strain was most closely related to Bacillus.According to the results of physiological and biochemical reactions,the strain was identified as Paenibacillus glucanolyticus.A series of experiments were carried out to study the physical and chemical properties of Paenibacillus glucanolyticus.It was found that Paenibacillus glucanolyticus grew well at the temperature between 30-60 ?,had certain bacteriostasis to common pathogenic bacteria,grew rapidly within 16-24 hours,and had the characteristics of weak acid resistance,bile salt resistance and heat resistance.Because endospores can be produced by Bacillus,the spores have thick walls and certain resistance to the external environment,it can survive in hot pure water,so it is hard to kill by conventional sterilization.In this paper,suspension quantitative disinfectant sterilization,acid potential water sterilization and high-pressure steam sterilization were compared.Finally,in practical application,high-efficiency disinfectant compound peracetic acid was used to kill the bacteria,and the killing effect was effectively monitored.
