Microbial flocculent(MBF)has a wide spread application prospect as a new type water treatment agent which is safe,efficient and can be natural degraded.And how to improve the output of microbial flocculent and reduce the production cost effectively has become a research hotspot.Paenibacillus sp.A9 CGMCC 2040 which has high flocculent production ability was screened from soil and the flocculent named MBFA9 has strong flocculation.The fermentation characteristics of Paenibacillus sp.A9,MBFA9,the flocculation and so on were studied in the research.The results of the study are as follows:(1)The conditions of fermentation were optimized by single factor experiment.And the results are:the dosing quantity of glucose which is carbon source is 15g/L,the shaking speed is 150r/min,the temperature is 30?,pH is 7 and the fermentation time is 24-36h.When the strain reaches a logarithmic growth,the glucose is consumed quickly and polysaccharide production increases,then the flocculation rate of 0.5%Kaolin reaches the highest level which is 97.83%.(2)Under the conditions of fermentation,DNS,spectrophotometry and kit method were used to determine the parameters changement and compare the metabolic difference of fermentation process which took Paenibacillus sp.A9 and two other strains as targets Paenibacillus Hunan Hunanenisis FeL05T(CGMCC 1.8907T)and Paenibacillus polymyxa(CGMCC1.4261T).The experiment showed that the time which three strains grow best are all 36h,but Paenibacillus sp.A9 and Hunanenisis FeL05T grow clearly better than P.polymyxa;pH levels off when it gradually reduced to 5.5;the production of polysaccharide of Paenibacillus sp.A9 is much more than the other two strains;the flocculation rate of Paenibacillus sp.A9 is greater than 95%after 36h and the other two are less than 50%;the content of ethanol dehydrogenase in fermented liquid is maximum at 36h which much more than the other two strains.All these showed that the metabolic process and production of glucose of Paenibacillus sp.A9 are different from the other two strains.(3)Origin8.0 was used to study the dynamics equations of bacteria growth,product synthesis and substrate consumption of Paenibacillus sp.A9 and the other two typical strains in fermentation process based on the model of Logistic,Luedeing-piret and substrate consumption and the fit of each equation are all above 97%.Each model can express changes of the corresponding item of strains in the process of fermentation well determined by the analysis of Excel.The study provided some technical parameters and theoretical basis for later mass of Paenibacillus sp.A9.
