Colorectal cancer(CRC)is a kind of malignancies occurring in the colon or rectum.It is the cancer with the second highest rate of mortality around the world.Screening biomarkers with good sensitivity and specificity is therefore propitious to the precision diagnosis of CRC,and it is also an effective way for monitoring the prognosis and treatment of CRC patients.This study can be divided into two parts.In the first part,we manually collected previously reported CRC epigenetic biomarkers and constructed the database called Epigenetic Biomarker Database for Colorectal Cancer(CRC-EBD,www.sysbio.org.cn/EBD).The database is in B/S structure so that the users can access it via the browser directly.Currently,a total of 334 items of epigenetic biomarkers were recorded in the database.In the second part,we integrated CRC methylation microarray data,protein-protein interactions and the records in CRC-EBD for the topological and functional characterization of reported CRC DNA methylation diagnostic biomarkers.Based on this observation,we further identified 23 potential methylation biomarkers.Among them,most CpGs had high AUC values and 10 genes were already been reported as candidate biomarkers or associated with CRC development.In addition,we performed functional enrichment analysis on the diagnostic biomarkers for systematically understanding of the relationship between the candidate biomarker genes and CRC carcinogenesis.Our database provides an interactive platform for doctors and researchers for the search and analysis of CRC epigenetics biomarkers.Meanwhile,the newly identified biomarkers based on network structural analysis may contribute to the diagnosis and treatment of CRC.
