This experiment combines the needs of the earthworm aquaculture industry.According to the living habits of the earthworms,it is found that in the cold regions,temperature is one of the abiotic stress factors that restrict the growth and reproduction of the earthworms.Today,the red seed loves earthworm species is the most widely cultivated species in the world.Earthworms belong to the genus Aisheng and are the most cultivated species in the alpine regions of China.The previous studies have been on physical and other external conditions and facilities.There is very little research on the temperature regulation of earthworms.In order to find a way to adjust the earthworms to adapt to the temperature,and observe the application in actual production,this experiment uses a parameter-free gene.The sequencing method compares the differential gene enrichment degree of the same species of earthworms under different breeding treatments to find the corresponding regulatory pathways.Due to the sweetness of earthworms,we selected different base materials for breeding comparison before inducing earthworm cold resistance and domestication,inducing the earthworms to feed,and selected the base material composition that can better allow the earthworm to overwinter;two groups of different In contrast,the base materials were all fermented cow manure,waste straw powder,sprayed water and molasses to maintain humidity,which laid the foundation for the subsequent domestication process and practical application.The main research contents and results are as follows:1?Cold resistance of different base materials on earthworms.The results show that the addition of molasses and straw powder to the earthworm base material can well increase the fat content of earthworms and help the earthworms to survive winter at low temperature.The addition of straw powder can adjust the carbon of the base material,which not only improves the base material.The breathability,and the effect of thermal insulation,but the effect is not significant.And through measurement,it was found that the fat content of earthworms cultured with molasses base was significantly higher than that of common earthworms,and the water content was higher than that of normal earthworms.2?Cold-resistant domestication of earthworms.According to the results of Experiment 1,normal base cultivation and molasses base cultivation were used,and domestication experiments were performed at the same time.The results showed that after 80 days of domestication,earthworms could eat at temperatures below 10 ?.Entering the early hibernation at 0?,the cold resistance of earthworms was significantly improved.This proves that the cold hardening domestication of earthworms was successful.3?Sequencing and analysis of cold-resistant genes related to earthworms.Through experiment two,the temperature of the earthworm's living environment was adjusted to allow the earthworms to breed under a cold-tolerant condition for a reproduction cycle.Transcriptome sequencing technology was used to detect earthworms cultured at normal temperatures and earthworms that had undergone cold hardening and acclimation to find differentially expressed genes.Enriched,GO analysis and KEGG analysis were performed,and the cold-resistant genes of earthworms were screened.The research results show that exploring cold-resistant genes of earthworms is a complex biological process,including metabolites,enzyme catalysis,and signal transduction.Wait.Earthworm cold resistance genes are regulated by external environmental conditions and internal co-regulation.4?Practical application of cold-resistant earthworms.In the northern high and cold regions,the temperature in winter can be as low as a dozen degrees below zero,and the cold resistance of domesticated earthworms has increased significantly.Through practical operations,it has been found that the survival rate of earthworms that have been reared in the cold in winter has been significantly improved.In addition,the temperature in the shed is increased by the sunlight transmitted through the greenhouse in the daytime.The dead leaves are laid in the middle and top when the earthworm base material is stacked,so that the dead leaves generate heat when they are fermented by the elevated temperature during the day,thereby increasing the base temperature.It is found that the breeding cycle of cold-resistant earthworms in this method has greatly improved the activity of earthworms and produced a small number of earthworm cocoons,which greatly reduced the cost of earthworm breeding,improved the breeding environment,and increased income.
