In order to preliminary understand the prevalence of Mycoplasma infection in large-scale sheep farms in part of Southern Xinjiang,and cultivate high-quality strains in Southern Xinjiang.During 2018~2019,132 nasal swabs of healthy and respiratory-stricken sheep and 6 lung tissue samples of dead lambs were randomly collected from 5 large-scale sheep farms in Kuqa,Yingjisha,and Bachu,southern Xinjiang.The samples were initially identified using PCR technology,some representative strains were selected for sequence analysis of their 16 S rRNA genes,and their molecular characteristics were analyzed using DNAStar,MAGE,and DNAMAN software,and a contiguous phylogenetic tree based on 16 S rRNA genes was constructed using the adjacency method;The mycoplasma liquid medium MTB and solid medium MTA were used to isolate and culture the pathogens.The colony morphology was observed with an optical microscope.The PCR method was used to determine the species at the molecular level.From 132 sheep nasal swabs and 6 lung tissue specimens from Bachu,Kuche and Ingesha,20 positive samples of Mycoplasma ovipneumoniae,16 positive samples of Mycoplasma arginine,4 positive samples of Mycoplasma conjunctiva,total positive The rate is 29.0%.The positive rates of 3 regions were 10.0%,50.0% and 15.4%,respectively.Phylogenetic tree analysis showed that the isolates and related strains belonged to the same large branch.Mycoplasma ovipneumoniae was closely related to the 2017-318-25 strain,Mycoplasma arginine was closer to E3.5 and G230 strains,and Mycoplasma conjunctiva was related to Goat 655 strains are closely related.Both Mycoplasma ovipneumoniae and Mycoplasma arginine do not belong to the same branch as domestic isolates.12 strains of Mycoplasma ovipneumoniae and 9 strains of Mycoplasma arginine were isolated from 40 PCR-positive samples.No Mycoplasma conjunctiva was isolated.16 S rRNA gene fragments and Mycoplasma-specific fragments were amplified as expected.Sequence analysis showed that Mycoplasma ovipneumoniae and Mycoplasma ovipneumoniae gene sequences published in GenBank shared 100% homology;Mycoplasma arginine and the sequence homology of Mycoplasma arginine gene published in GenBank is 100%.
