ObjectiveThe flies,as Musca domestica,M.domestica vicina,Drosophilia melanogasterhas etc.considered potential agents for disease transmission ever since they are existence.The habitual movement of house flies from filthy substrata such as human faeces,animal excreta,carcasses,garbage,etc.makes them ideal candidates for disease transmission.The objective of this study was to study the microbial fauna isolated from the external surfaces of the fly samples,and assess the pathogen transmission potential under the lab environment.Methods1.The microbial fauna isolated from the external surfaces of the fly samples.The adult flies were captured with the sweep net,and use the normal isolation technique to isolate the bacteria.The solutions were then spread evenly on the agar plates.All the bacteria were identified to genus level by 16 S rDNA pyrosequencing.2.Bacterial diversity from the external surfaces of the fly samples captured in farms.The total DNA of the samples were used Illumina MiSeq to sequence by sequencing company.3.The pathogen transmission potential under the lab environment.The laboratory-bred adult house flies were put in the agar plates containing Escherichia coli for 30-90 min,and then examine the concentration of the badteria obtained from the external surfaces of the adults at different time interva.Results1.The microbial fauna isolated from the external surfaces of the fly samples.Musca domestica,were the dominant specie collected from all collection sites.Castellaniella,Paracoccus,Hafnia,Gluconobacter,Comamonas,Psychrobacter,Acidiphili um,Agrobacterium,Pediococcus,Kocuria,Advenella,Leuconostoc,Rhodococcus,lactococcus,Corynebacterium,Ochrobactrum,Arthrobacter,Lysinibacillus,Stenotrophomonas,Microb acterium,Enterobacter,Bacillus,Klebsiella,Staphylococcus,Proteus,Escherichia,Serratia,myroides,Providencia,Acinetobacter,Weissella,Enterococcus,Streptomyces,Pseudomonas,Raoultella,Flavobacterium,Citobacter,Alcaligenes,Sphingobacterium,Pectobacterium,Pa ntoea,Brachybacterium,Shigella and Azospirillum were isolated from external surfaces of house fly.2.Bacterial diversity from the external surfaces of the fly samples captured in farms.Using 16 S rDNA pyrosequencing,we obtained 966633 PCR products from the different livestock farms.The combined diversity of bacteria classified across livestock farms spanned sixteen bacterial phyla,Proteobacteria,Firmicutes,Actinobacteria and Bacteroidetes were the most dominant phyla accounting for 99.5% of the fauna identified.Of the bacterial genera identified,Proteus were the most dominant genera in YNC-1-LC and YYC,Klebsiella were the most dominant genera in YNC-1-NS,Bacteroides were the most dominant genera in YZC-1,Kurthia were the most dominant genera in YZC-2.3.The pathogen transmission potential under the lab environment.After that interval processing time for 12 hours,The bacterial concentrations of three experimental group were reduced to the initial concentration of 2 / 100,2 / 100,100.After24 hours were reduced to the initial concentration of about 1/1000.Conclusion1.The house flies are the main species of Musca domestica in garbage and farm.2.The house fly breed in polluted environment have higher incidence to transport pathogens.3.The flora structure of different environment is diverse,and mainly related to the ecological types,geographic location.
