Since the start of the Human Genome project,multiplexed assay has become a hot area of research.Planar arrays have many disadvantages,so suspension arrays which are based on encoded microcarriers become one of the most promising high-throughput multiplexed assay technique.Quantum dots(QDs)with superior multiplexing capability are excellent for the encoded microspheres.At the same time,the decoding strategies based on the flow cytometry become the main test and analysis platform,with the advantage of convenience,high speed and high throughput.The purpose of this paper is to explore the scientific law between the preparation of QDs encoded beads and their decoding by flow cytometry.Firstly,four dual QDs encoding modes were designed.Then,based on the method of PSMA-mediating QDs and the method of modified LBL,six type of encoded beads were prepared which were used to study the distribution of each cluster by flow cytometry.It indicates that the controllability of PSMA-mediating QDs and the method of modified LBL is high,and the condition of the preparation process has some influence on the distribution of each cluster tested by the flow cytometry.After that,the typical three QDloading modes were selected to prepare multiple encoded beads.20 clusters were tested by flow cytometry under each coding mode,so the changing rule and the influencing factors of the center of the multiple clusters could be found.The results show that the difference between the coding modes has some influence on the coding array tested by the flow cytometry.Finally,the mode which was most conducive to achieve high-throughput and precise coding of QDs was chosen to study the loading mechanism by exploring the loading characteristics on the surface of the microspheres of the two kind of QDs respectively.It shows the relationship between the coding array and the preparation of the encoded beads.This study provides a theoretical guidance for the preparation of QDs encoded beads with ultrahigh coding capacity and high accuracy.
