| Mileewinae belongs to Hemiptera,Cicadellidae.At present,there are 166 valid species(subspecies)of 4 tribes and 8 genera in the subfamily Mileewinae all over the world.It is widely distributed in the Oriental boundary,the African boundary and the neotropical boundary,with a few distributed in the palaearctic boundary.And now,a total of 60 species,1 tribe,3 genera of Mileewinae were recorded in China.In this paper,36 species of Mileewinae belonging to 3 genera were used as the phylogeny research objects,and the gene fragments of COI,16 SrDNA,28SrDNA,ITS2,COII and Cytb were preliminarily screened.Finally,COI,16 SrDNA and Cytb were selected for phylogenetic analysis of the insects of the subfamily Mileewinae in China.At the same time,we preliminarily obtained the mitochondrial genome sequence information of Mileewa albovittata and conducted phylogenetic analysis with the mitochondrial genome sequences of the representative species of the subfamily of leafhopper.The results are as follows:A total of 17 COI sequences from 2 genra,17 16 S rDNA sequences from 2 genra,16 Cytb sequences from 2 genra were obtained in this experiment.Phylogenetic analysis software like MEGA X and Mrbayes3.2.6 were used to construct 20 phylogenetic trees by using NJ,MP,ML and BI methods.Based on 20 phylogentic trees constructed by two software and multiple methods,the following conclusions are drawn:(1)Mileewa cockiheada + Mileewa ussurica +Mileewa alara,Ujna maolanana + Ujna puerana + Mileewa lackstripa + Mileewa gaoligongana,Mileewa branchiuma + Mileewa amplimacula + Mileewa margheritae has the most stable topology.(2)Ujna maolanana and Ujna puerana,Mileewa cockiheada and Mileewa ussurica,Mileewa mira and Mileewa ponta,Mileewa amplimacula and Mileewa margheritae,Mileewa coeomacula and Mileewa branchiuma all have the sister relationship each other.(3)The results support genus Processina as monophyletic group while the monophyletism of Ujna was not supported.The circular mitogenome of M.albovittata is 15,079 bp in size,containing 13 protein-coding genes(PCGs),22 tRNA genes,two rRNA genes,and one AT-rich region,The A+T content is 79.6%.Among them,total length of 13 protein-coding genes(PCGs)is 10,965 bp,encoding 3640 amino acids.Eleven PCGs have ATN as the start codon,except for ATP8 and NAD5 genes have TTG.Twelve PCGs have TAA as the stop codon,while COII gene has an incomplete stop codon(A).The LSU(16S)and SSU(12S)of rRNA gene are 1,220 bp and 749 bp.BI tree was constructed based on PCGs of mitochondrial genome of Mileewa albovittata and 19 species of GenBank published cicadellidae insects,we can find that M.albovittata,Empoasca onukii and Empoasca vitis have the sister relationship each other.And M.albovittata was also close to two species of subfamily Cicadellinae. |
