long noncoding RNA(lncRNA)is defined as non-protein coding transcriptes longer than 200 nucleotides.More and more studies indicate that lncRNA not only plays an important role in normal physiological processes of cells but also has a great effect on governing cancer initiation and development.It is one of hot points that how lncRNA and the networks related with it regulate the tumor.The regulator of reprogramming(RoR)is a kind of lncRNAs that has recently been identified.The lieratures have pointed out that lncRNA RoRplay a negative role in regulating the expression of tumor suppressor gene p53.But the dynamics is still unclear.The project is conducted using experiment and simulation to study the dynamics mechanism.The valuable results are obtained as follows:The experimental results indicate: The expressions of lncRNA RoR and p53 mRNA show the same change trendin human breast cancer cells induced bydoxorubicin;the experiment confirmed againthe negative feedback mechanism between lncRNARoR and p53.lncRNA RoR can inhibit the expression of P53 protein and P53 proteincan promote lncRNA RoR expression;The oscillation periods of lncRNA RoR is 270±30 min,the oscillation periods of P53 is 195 ±15 min,which is agree with the results in literature.The dynamical equations are first proposed.The simulation results by solving the equations are agreeable to the experimental results at given doxorubicin intenstiy.The expresions of lncRNA RoR and P53 increase with the intensity of doxorubicin.The dynamical relationship between lncRNA RoR and P53 is sensitive to the time that doxorubicin is injected.
