Medicago falcata L.belongs to the Medicago,and it is a kind of leguminous herbage with wide distribution,strong resistance,diverse morphology,rich nutritional value.In order to study the genes of Medicago falcata L.that can respond to both biotic and abiotic stresses,this paper covers the study is that the MfERF049 gene of wild Medicago falcata L.was obtained by the technology of molecular biology and transformed into the root of Madicago truncatula A17.The function of MfERF049 gene in improving plant stress resistance was analyzed.In the early stage,Medicago falcata L.was treated with salt and low temperature stress in the laboratory,and the genes with significant differences in expression were screened out from the transcriptome sequencing results.On this basis,the bioinformatics analysis in this study revealed that the c70245-g2 sequence is homologous gene of MtDREB2 A in Medicago truncatula A17.The specific research methods are as follows: Firstly,specific primers were designed using the known MtDREB2 A gene sequence as template,and analyzed the expression pattern of the target gene MfERF049 with wild Medicago falcata L.under different conditions.Then the MfERF049 gene from Medicago falcata L.was cloned by the method of homologous gene cloning.Then,MfERF049 was constructed into the expression vector pKGWRR,and transformed into the root of Madicago truncatula A17 by the method of hairy root transformation method.The results of phenotype(root length and root fresh weight)of the transgenic root was statistically analyzed,and the results of the physiological indicators(POS,SOS,MDA and PRO)detection and the expression levels of the target genes and its downstream genes of the transgenic roots were analyzed.The study demonstrated that the MfERF049 gene responded to high salt,drought,high temperature,low temperature,ABA and Sinorhizobium stress by stress experiments,especially high salt,drought and low temperature stress,which is consistent with the research results of this gene in other species included inland and abroad.In addition,the MfERF049 gene can positively regulate Lea14 and negatively regulate Cas15 A,respectively.In this paper,homologous cloning method was used to obtain the sequence of MfERF049 gene of wild Medicago falcata L.and preliminarily explore the function of this gene in the resistance of plant,deepening the understanding of AP2/EREBP family.
