Rop, a plant-specific small GTPases, is a subfamily of RHO GTPases. ROPs are also involved in the development of cell polarity in pollen tubes and the recent studies demonstrate a pivotal role for ROPs in regulating various aspects of root hair development.In the development of root hair the rhizobium-legume interactions depend on the form of the infection thread. The form of the infection thread is a polarity development .The research aim of this study is to obtain the genes regulating polarity developing of the infection thread in the rhizobium-legume interactions to provide some information about the function of the ROP GTPase between symbiotic partners.Using biological informatics and homology gene clone methed ,with the technique of RT-PCR,two cDNA colones from Medicago truncatula,ROP2 and ROP6,were obtained.The full-length of the ROP2 and ROP6 containing a 594bp ORF,which encodes 197 amino acids. The deduced amino acid sequence alignment shows that ROP2 and ROP6 have >50% identity with the 11ROPs from Arabidopsis thaliana ;According to the expression pattern ,the expression of the two genes is not tissure-specific ,but the two gene induced by rhizobium expressed strongly to some extent;The results showed that plant-specific ROP proteins maybe play an important role in the rhizobium-legume interactions.We constructed the plant expressinon vectors of ROP2 and ROP6 and transformed them into Medicago truncatula.
